“温阳通脉”灸法对动脉粥样硬化小鼠血脂及肝脏CPT1α、Mcad含量的影响

The Effect of “Warming Yang and Freeing Vessel” Moxibustion on Blood Lipids and the Contents of CPT1α and Mcad in the Liver of Atherosclerotic Mice

摘要 目的 探讨“温阳通脉”灸法对ApoE^(-/-)动脉粥样硬化(AS)小鼠血脂及肝脏肉毒碱棕榈酰转移酶1α(CPT1α)、中链脂酰CoA脱氢酶(Mcad)含量的影响。方法 选取普通饲料喂养的10只C57BL/6J小鼠为对照组,高脂饲料喂养的20只ApoE^(-/-)小鼠为模型组和艾灸组。艾灸组取小鼠“膻中”“神阙”以及双侧的“内关”“血海”处行艾灸疗法。实验结束后,采用全自动生化分析仪,分别检测小鼠的血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)及高密度脂蛋白胆固醇(HDL-C)的含量;HE染色观察各组小鼠主动脉弓及肝脏病理形态;油红O染色观察各组小鼠肝脏脂质沉积;ELLSA法检测肝脏中CPT1α、Mcad含量。结果 与对照组比较,模型组小鼠在第12周体质量明显增加(P<0.01),血清中TC、TG、LDL-C含量均明显升高(P<0.01),HDL-C含量明显降低(P<0.05),肝脏CPT1α、Mcad表达量降低(P<0.01);HE与油红O染色显示主动脉血管内膜增厚粗糙,有泡沫细胞沉积,肝细胞形态异常,脂肪变性,脂质蓄积显著增加。与模型组相比,艾灸组小鼠第12周体质量显著降低(P<0.01),血清TC、TG、LDL-C含量均显著降低(P<0.01),肝脏CPT1α、Mcad表达量增加(P<0.01);HE与油红O染色显示主动脉血管内膜无明显斑块形成,但部分内皮细胞肿胀变性,肝细胞形态改善,脂质蓄积显著减少。结论 “温阳通脉”灸法可调节AS小鼠的脂质代谢,改善主动脉组织病理损伤,其潜在机制可能与增加肝脏组织中CPT1α、Macd的表达,促进脂肪酸氧化,减少肝脏脂质沉积相关。 Objective To investigate the effects of moxibustion method"Warming Yang and Freeing Ves-sel"on blood lipids and liver carnitine palmitoyltransferase 1 alpha(CPTlα)and medium-chain fatty acyl CoA dehydrogenase(Mcad)contents in ApoE-/-atherosclerotic(AS)mice.Methods Ten C57BL/6J mice fed with regular feed were selected as the control group,while 20 ApoE-'-mice fed with high-fat feed were selected as the model group and the moxibustion group.In the moxibustion group,moxibustion was applied to the"Dan Zhong","Shen Que"and bilateral"Nei Guan"and"Xue Hai"areas on the mice.After the experiment,a fully automated biochemical analyzer was used to measure the levels of serum total cholesterol(TC),triglycer-ides(TG),low-density lipoprotein cholesterol(LDL-C),and high-density lipoprotein cholesterol(HDL-C)in the mice,respectively.HE staining was performed to observe the pathological morphology of the aortic arch and liver in mice of each group;Oil Red O staining was used to observe lipid deposition in the liver of mice in each group;and the ELISA method was employed to detect the levels of CPTla and Mcad in the liver.Results Compared with the control group,the body mass of mice in the model group was remarkably increased at week 12(P<0.01).The levels of serum total TC,TG,LDL-C all were remarkably elevated(P<0.01).The content of HDL-C was remarkably decreased(P<0.05),and the expression levels of hepatic CPT1αand Mcad decreased(P<0.01).HE and oil red O stain showed that the intima-media of the aortic vasculature was thick-ened,the plaque formation,abnormal hepatocyte morphology,steatosis,and remarkable increase in lipid accu-mulation.Compared with the model group,mice in the moxibustion group showed a remarkable decrease in body mass at week 12(P<0.01),a remarkable decrease in the levels of serum TC,TG and LDL-C content(P<0.01),and an increase in the expression levels of hepatic CPTlαand Mcad expression(P<0.01).HE and oil red O stain showed that there was no significant plaque formation in the intima-media of the aorta vessels,but some of the endothelial cells were swollen and degenerated,and the morphology of the hepatocytes was im-proved,with a significant decrease in lipid accumulation.Lipid accumulation was significantly reduced.Conclu-sion"Warming Yang and Freeing Vessel"Moxibustion method can regulate lipid metabolism and improve the pathological damage of aortic tissues in AS mice.Its potential mechanism may be related to the increase of the expression of CPTlαand Macd in liver tissues,the promotion of fatty acid oxidation,and the reduction of he-patic lipid deposition.
