摘要
目的:通过牙周临床检查与16S rRNA高通量测序技术,对非阻生第三磨牙存在时第一磨牙和第二磨牙的牙周状况以及龈下菌斑微生态进行对比研究。方法:依据纳入和排除标准选取观察对象,清除局部龈上菌斑,开展必要的口腔卫生教育。对观察区域的第一磨牙和第二磨牙进行牙周探诊检查,同时采集局部龈下菌斑,利用PacBio平台进行全长16S rRNA测序,对比两者的牙周临床指标和局部微生物群落特征。结果:与非阻生第三磨牙相邻时,第二磨牙平均探诊深度、探诊出血百分比、深牙周袋比率和菌斑指数均显著高于第一磨牙;第二磨牙龈下菌斑微生物的多样性和丰富度明显大于第一磨牙,β多样性对比分析表明两组间微生物群落结构存在显著差异。在第二磨牙龈下菌斑中,牙龈卟啉单胞菌、具核梭杆菌等牙周致病菌的相对丰度显著高于第一磨牙。结论:与非阻生第三磨牙相邻时,第二磨牙的牙周状况及龈下菌斑微生态较第一磨牙差。
AIM:To compare the periodontal status and subgingival plaque microecology of the first and second molars when non-impacted third molars were present,through periodontal clinical examination and 16S rRNA highthroughput sequencing technology.METHODS:Subjects were included according to the inclusion and exclusion criteria.Oral hygiene instruction and supragingival plaque removal were performed.Subsequently,periodontal probing examination was conducted on the first and second molars of the observed regions,and the local subgingival plaque was collected.Full-Length 16S rRNA Sequencing was carried out using the PacBio platform.Finally,compared the periodontal clinical indicators and local microbial community characteristics of the first and second molars.RESULTS:The average probing depth,percentage of bleeding on probing,percentage of deep periodontal pockets,and plaque index of the second molars adjacent to nonimpacted third molars were significantly higher than those of the first molars.The diversity and richness of subgingival plaque microorganisms of the second molars were significantly greater than those of the first molars.β-diversity analysis showed significant differences in the microbial community structure between the two groups.In the subgingival plaque of the second molars,the relative abundances of periodontal pathogens such as Porphyromonas gingivalis and Fusobacterium nucleatum were significantly higher than those of the first molars.CONCLUSION:When adjacent to non-impacted third molars,the periodontal status and subgingival plaque microecology of the second molars are worse than that of the first molars.
