志不安于肾不寐模型大鼠听觉区脑组织凋亡因子表达研究

摘要 目的通过检测志不安于肾不寐模型大鼠听觉区脑组织及肾组织Bcl-2、Bax、Caspase-3和NF-κB表达水平,探讨志不安于肾不寐的发病机制。方法SPF级雄性SD大鼠16只,随机分为正常组和模型组,各8只。模型组采用颈背部皮下注射D-半乳糖联合腹腔注射对氯苯丙氨酸制备志不安于肾不寐模型大鼠;正常组给予等剂量生理盐水注射。用HE染色观察2组大鼠肾组织病理变化,免疫组化及Western blot检测2组大鼠相关脏器中Bcl-2、Bax、Caspase-3、NF-κB的表达水平。结果与正常组比较,模型组大鼠终体质量明显减轻(P<0.001);模型组大鼠睡眠潜伏期显著延长及睡眠时间显著缩短(P<0.001);模型组在目标象限滞留时间增加,进入目标象限次数增加,在目标象限运动距离增加,最终找到目标靶平台运动轨迹最长(P<0.05);模型组大鼠肾HE染色示肾小球及肾小球结构紊乱;免疫组化结果示模型组大鼠听觉区脑组织及肾组织Bcl-2表达减少,Bcl-2/Bax比值降低(P<0.05),Bax、Caspase-3、NF-κB表达增加(P<0.05);Western blot结果示模型组大鼠肾组织和听觉区脑组织Bcl-2减少(P<0.05),NF-κB、Caspase-3、Bax显著增加(P<0.05)。结论脑组织及肾组织凋亡水平的改变与志不安于肾不寐的发生有关。 Objective To investigate the pathogenesis of insomnia by detecting the protein expression levels of Bcl-2,Bax,Caspase-3 and NF-κB in the brain tissue of the auditory region and the kidney tissue of rats with insomnia caused by the kidney failing to store the will.Methods A total of 16 SPF male SD rats were randomly divided into a control group(n=8)and a model group(n=8).In the model group,D-galactose was injected subcutaneously in the back of the rat neck and p-chlorophenylalanine was injected intraperitoneally to prepare a model of rats with insomnia caused by the kidney failing to store the will,while the control group was injected with the same dose of normal saline.HE staining was used to observe the pathological changes in renal tissue of the two groups,and immunohistochemistry(IHC)and WB were used to detect the expression levels of Bcl-2,Bax,Caspase-3 and NF-κB in the relevant organs of the two rat groups.Results Compared with the control group,the terminal body weight was significantly reduced in the model group(P<0.001),and the sleep latency was significantly prolonged,yet the sleep time was significantly shortened(P<0.001),while the residence time in the target quadrant was increased,the number of times to enter the target quadrant was increased,and the movement distance in the target quadrant was increased,and the target platform was found to have the longest trajectory(P<0.05).HE staining of the kidney in the model group showed disorders in the glomeruli and glomerular structure,and the IHC results showed that the expression of Bcl-2 in brain tissue in the auditory area and the kidney tissue of the model group was decreased,and the ratio of Bcl-2/Bax was decreased(P<0.05),while the expression levels of Bax,Caspase-3 and NF-κB were increased(P<0.05).The WB results showed that the expression of Bcl-2 in the kidney tissue and brain tissue of the auditory area of the model group was decreased(P<0.05),while the expression levels of NF-κB,Caspase-3 and Bax were increased significantly(P<0.05).Conclusion The changes in the level of apoptosis in the brain tissue and kidney tissue are correlated with the occurrence of insomnia caused by the kidney failing to store the will.
