摘要
为了明确菌株HY-021生长的最适条件,对其发酵产孢条件的优化进行探索。本文采用菌丝生长速率法和孢子数测定法,对拟蚕豆葡萄孢(Botrytis fabiopsis)HY-021菌株除草活性在不同培养条件下的生长及产孢情况进行了研究。结果表明,PSA培养基最适合HY-021菌株菌丝生长和产孢,最适温度为20℃,以肌醇为碳源,硫酸铵为氮源且在黑暗条件下有利于菌丝生长,最适无机盐为氯化钙,菌丝生长的最适pH值为6~9,产孢的最适pH值为7~8,过酸过碱对HY-021菌株菌丝生长及产孢均不利。由发酵条件优化得出,HY-021菌株最佳发酵时间为5 d,最适接种量和装液量为3%和160 mL,最佳摇床转速为180 r/min。这将为菌种的大规模生产与剂型研发提供参考。
In order to clarify the optimum conditions for the growth of strain HY-021,the optimization of its fermentation and spore production conditions was explored.In this paper,the growth and spore production of Botrytis fabiopsis strain HY-021 under different culture conditions were investigated by using the mycelial growth rate method and spore number determination.The results showed that PSA medium was the most suitable for mycelial growth and spore production of HY-021 strain,the optimum temperature was 20℃,inositol was used as the carbon source,ammonium sulfate was used as the nitrogen source and it was favorable for mycelial growth in the dark,the optimum inorganic salt was calcium chloride,the optimum pH for mycelial growth was 6~9,and that for spore production was 7~8,and too acidic and too alkaline were unfavorable to the growth of mycelia and spore production of HY-021 strain.From the optimization of fermentation conditions,the optimal fermentation time for HY-021 strain was 5 d,the optimal inoculum and liquid loading were 3%and 160 mL,and the optimal shaker speed was 180 r/min,which will provide a reference for the large-scale production of strains and the development of dosage forms.
