摘要
明确了广东省阳春市花滩林场土沉香(Aquilaria sinensis)根围土壤中的针线虫种类.采用形态特征、形态测量值和rDNA分子特征(28S D2D3区和ITS区)以及基于两个rDNA分子片段的系统发育分析对针线虫种群实施系统鉴定.广东省土沉香针线虫种群形态特征与原始文献中的拟隐针线虫(Paratylenchus paralatescens)一致,仅雌虫尾末端形态有差异.广东种群雌虫主要形态特征:侧区具3条侧线,口针长76.8-79.3μm,唇区前端平截,排泄孔位于口针基部球之前.阴门唇不突出,且没有阴门膜;受精囊大,近似长方形,内充满精子.尾长24.4-28.8μm,纤细且比较直,末端圆.拟隐针线虫广东种群28S rDNA D2D3区和rDNA-ITS区与GenBank数据库的浙江种群进行序列比对,相似性分别为99.4%和99.7%.贝叶斯推断法构建的系统进化树显示:拟隐针线虫广东种群与原始报道的浙江种群聚为高度支持的分支.结合形态学观察和分子序列分析(28S rDNA D2D3区和rDNA-ITS区),将广东省土沉香针线虫种群鉴定为拟隐针线虫.拟隐针线虫是广东省植物线虫新纪录种.土沉香是拟隐针线虫的新寄主.
This study is to clarify the species of pin nematode isolated from rhizosphere soil of Aquilaria sinensis at Huatan forest farm in Yangchun City,Guangdong Province.Species identification of the pin nematode was performed by morphology,morphometrics and molecular characterization of two rDNA(28S D2D3 and ITS)sequences,as well as phylogenetic analyses of the two rDNA sequences.The morphological characteristics of the pin nematode population from A.sinensis in Guangdong was in according with those of Paratylenchus paralatescens in original paper except for the female tail terminus shape.It was characterized by the female lateral field with three lines,stylet 76.8 to 79.3μm long,lip region truncated,excretory pore located anterior to basal knobs.Vulval lips are not protruded and without vulval flap;spermatheca large,rectangular-shaped and filled with sperms.Tail 24.4 to 28.8μm long,slender and relatively straight,having round shape terminus.28S rDNA D2D3 and rDNAITS sequences of P.paralatescens Guangdong poputation showed high similarities(99.4%and 99.7%,respectively)with the sequences of Zhejiang population.Bayesian inference(BI)trees inferred from the two rDNA sequences revealed that P.paralatescens of this study formed a highly supported clade with the original Zhejiang population.The pin nematode in this study was identified as P.paralatescens by morphological characteristics and molecular analyses.P.paralatescens is a new record species of pin nematode in Guangdong Province.A.sinensis is a new host record of P.paralatescens.
