摘要
为了探讨果胶寡糖抑制人结肠癌细胞DLD-1的增殖作用及其相关机制,采用CCK-8法检测果胶寡糖对细胞增殖的抑制作用,流式细胞仪检测果胶寡糖诱导结肠癌细胞的凋亡情况,RT-PCR方法检测细胞中抗凋亡基因表达,Western blot方法检测细胞中抗凋亡蛋白(Bcl-2)和促凋亡蛋白(Bax和Caspase-3)的表达。实验结果标明:浓度为1000μg·mL^(-1)的果胶寡糖处理DLD-1细胞48 h后,细胞活力下降到61%,细胞凋亡率为31.67%;果胶寡糖通过降低Bcl-2蛋白表达水平,升高Bax蛋白的表达水平,提高Bax/Bcl-2比率,激活Caspase-3蛋白诱导DLD-1细胞的凋亡,发挥抑制结肠癌细胞的作用。为开发新型抗结肠癌药物提供一定理论依据。
To investigate the inhibitory effect of pectic oligosaccharides on the proliferation of human colorectal adenocarcinoma(DLD-1 cells)and its underlying mechanisms in this study,CCK-8 assay was used for evaluation of proliferation of DLD-1,and flow cytometry was utilized to detect the apoptotic cells.RT-PCR was used to detect the expression of anti-apoptotic genes in cells,while Western blot was adopted to detect the expression of anti-apoptotic proteins(Bcl-2)and pro-apoptotic proteins(Bax and Caspase-3)in cells.The results demonstrated that after treatment of DLD-1 cells with 1000μg·mL^(-1) for 48 h,the cell viability decreased to 61%,and the apoptosis rate was 31.67%by flow cytometry assays.Pectin oligosaccharides inhibited DLD-1 cells by reducing the expression level of Bcl-2,increasing the expression level of Bax,increasing the ratio of Bax/Bcl-2,and activating Caspase-3 to induce apoptosis of tumor cells.This study provides theoretical basis for developing new anti-colon cancer drugs.
