摘要
目的:采用UPLC-Q-TOF/MS分析仪器,利用非靶向血清代谢组学技术分析参地补肾胶囊对肾间质纤维化小鼠血清的干预机制。方法:选用30只6~8周龄雄性C57BL/6J小鼠,随机分为假手术组(Sham组)、单侧输尿管结扎组(UUO组)、参地补肾胶囊组(SDBS组),每组10只。预给药1周后行单侧输尿管梗阻手术,术后继续给药3周后处死小鼠,收集小鼠肾脏、血清。分别进行HE、Masson染色评估肾脏组织病理改变,并通过对代谢物及其代谢通路的分析进一步探讨参地补肾胶囊对UUO小鼠血清代谢变化的干预机制。结果:经肾脏组织病理评估发现参地补肾胶囊对UUO小鼠肾脏组织形态具有明显改善效果。经代谢组学分析发现,参地补肾胶囊主要通过回调3条通路(甘油磷脂代谢、醚脂代谢、柠檬酸循环)及与其相关的14种代谢物对肾间质纤维化发挥干预作用。结论:参地补肾胶囊可有效改善UUO小鼠肾脏病理改变及代谢变化,改善小鼠脂质紊乱,达到干预肾间质纤维化、减轻肾损伤的作用。
Objective:To analyze the mechanism of action of Shendi Busen Capsule on serum metabolism in mice with renal interstitial fibrosis using UPLC-Q-TOF/MS and non-targeted metabolomics.Methods:Thirty 6-8 week-old male C57BL/6J mice were randomly divided into three groups:sham-operated group(Sham),unilateral ureteral obstruction group(UUO),and Shendi Busen Capsule group(SDBS).After pre-treatment for one week,unilateral ureteral obstruction surgery was performed,followed by continuous administration for 3 weeks before euthanizing the mice.Kidney tissues and serum were collected.HE and Masson staining were used to assess renal tissue pathological changes.The intervention mechanism of Shendi Busen Capsule on serum metabolic changes in UUO mice was further explored through analysis of metabolites and metabolic pathways.Results:Histopathological evaluation of kidney tissues revealed that Shendi Busen Capsule significantly improved renal morphology in UUO mice.Metabolomics analysis showed that Shendi Busen Capsule primarily intervened in renal interstitial fibrosis by modulating three pathways(glycerophospholipid metabolism,ether lipid metabolism,and the tricarboxylic acid cycle)and 14 related metabolites.Conclusion:Shendi Busen Capsule effectively improves kidney pathological changes and metabolic alterations in UUO mice,alleviates lipid disorders,and intervenes in renal interstitial fibrosis,thus reducing renal injury.
