摘要
目的:研究不同剂量环磷酰胺(CTX)治疗A549人源肺癌细胞荷瘤裸鼠时对免疫系统与肿瘤生长的影响。方法:将A549人源肺癌细胞接种于BALB/c裸鼠皮下,建立裸鼠荷瘤模型,按照肿瘤的体积随机划分为模型组、CTX低剂组、CTX高剂组,5-Fu+CDDP中剂量组设为阳性对照组,8只/组。另设空白对照组8只不接种肿瘤,每天注射对应的药物,模型组给予等量的生理盐水。每周称量动物体重、肿瘤体积两次。实验结束后,测量肿瘤和脾脏重量,ELISA法检测血液中IFN-γ,IL-2,IL-12,和IL-10的表达,并使用流式细胞仪检测脾脏组织中T淋巴细胞亚群中CD4+、CD8+的表达。结果:在测定过程中,CTX高剂量、CTX低剂量组在各测定时间点的体质量降低均无显著性差异(P>0.05),但CTX高剂量组的抑瘤率明显高于低剂量组,且5-Fu+CDDP中剂量组在给药过程中部分裸鼠出现消瘦、厌食、精神紧张等状况相对高、低剂量较为明显;在外周血中IL-2、IL-10、IL-12和IFN-γ含量情况检测中,CTX低剂量组、CTX高剂量组小鼠血清中INF-γ质量浓度明显下降(P<0.05),其中CTX高剂量甚至十分显著性差异(P<0.01),另外CTX高剂量组、5-Fu+CDDP中剂量组小鼠血清中IL-10质量浓度明显下降(P<0.05);脾脏组织中T淋巴细胞亚群中CD4+、CD8+的表达中,CTX高剂量组脾脏中CD4+CD8-T细胞数量明显增加(P<0.05),此外,CD4/CD8有十分显著性差异(P<0.01)。结论:CTX可能出现增强裸鼠的免疫调节能力,从而增强裸鼠脾脏的抗肿瘤免疫的作用;磷酰胺高剂量在给药组中较为适合。
Objective:To study the effects of different doses of cyclophosphamide(CTX)on the immune system and tumor growth in nude mice bearing A549 human lung cancer cells.Methods:A549 human lung cancer cells were inoculated subcutaneously into BALB/c nude mice to establish a nude mouse tumor-bearing model.The mice were randomly divided into a model group,a low-dose CTX group,a high-dose CTX group according to the tumor volume.The 5-Fu+CDDP medium-dose group was set as the positive control group,with 8 mice in each group.In addition,8 mice in the blank control group were not inoculated with tumors.The corresponding drugs were injected daily,and the model group was given an equal amount of normal saline.The body weight and tumor volume of the animals were weighed twice a week.After the experiment,the weights of tumors and spleens were measured.The expressions of IFN-γ,IL-2,IL-12,and IL-10 in the blood were detected by ELISA,and the expressions of CD4+and CD8+in T lymphocyte subsets in spleen tissues were detected by flow cytometry.Results:During the measurement process,there was no significant difference in the decrease of body weight at each measurement time point between the high-dose CTX group and the low-dose CTX group(P>0.05).However,the tumor inhibition rate of the high-dose CTX group was significantly higher than that of the low-dose group.And in the 5-Fu+CDDP medium-dose group,some nude mice showed more obvious conditions such as emaciation,anorexia,and nervousness during the drug-administration process compared with the high-and low-dose groups.In the detection of the contents of IL-2,IL-10,IL-12,and IFN-γin peripheral blood,the mass concentration of INF-γin the serum of mice in the low-dose CTX group and the high-dose CTX group decreased significantly(P<0.05),and the decrease in the high-dose CTX group was even very significant(P<0.01).In addition,the mass concentration of IL-10 in the serum of mice in the high-dose CTX group and the 5-Fu+CDDP medium-dose group decreased significantly(P<0.05).In the expressions of CD4+and CD8+in T lymphocyte subsets in spleen tissues,the number of CD4+CD8-T cells in the spleen of the high-dose CTX group increased significantly(P<0.05).In addition,there was a very significant difference in CD4/CD8(P<0.01).Conclusion:CTX may enhance the immune-regulating ability of nude mice,thereby enhancing the anti-tumor immunity of the spleens of nude mice.The high-dose of cyclophosphamide is more suitable in the drug-administration groups.
