高效液相色谱法测定四种降糖药物的研究

摘要 目的目的建立测定苯甲酸阿格列汀、磷酸西格列汀、维格列汀、利拉利汀四种二肽基肽酶Ⅳ抑制剂降糖药物的高效液相分析方法。方法方法采用waters色谱柱X-Bridge C18(250mm×4.6mm×5µm);流动相组成:流动相A为0.02mol/L醋酸铵溶液,流动相B为乙腈,采用梯度洗脱;检测波长为210nm;柱温30.0℃;进样量20µL。结果结果四种二肽基肽酶Ⅳ抑制剂降糖药物完全分离,检出限在0.10-3µg/ml之间;具有较宽的线性范围和良好的线性关系;日内、日间RSD分别为0.04%-1.08%之间和0.91%-5.36%之间;加样回收率均大于75%;RSD为0.04%-5.16%。结论结论该方法可用于四种化学降糖药物检测。 Objective A high performance liquid chromatography(HPLC)method was established for the determination of four dipeptidyl peptidase IV inhibitors antidiabetic drugs,alogliptin benzoate,sitagliptin phosphate,vildagliptin,linagliptin.Methods The separation was performed on a waters column X-Bridge C18(250mm×4.6mm×5µm),Mobile phase composition:Mobile phase A is 0.02mol/L ammonium acetate solution,mobile phase B is acetonitrile,using gradient elution.The detection wavelength was 210nm,the column temperature was 30.0℃,and the injection volume was 20µl.Results Four dipeptidyl peptidase IV inhibitors were separated completely,and the detection limit was between 0.10-3µg/ml.They had a wide linear range and good linear relationship.The intra-day and inter-day relative standard deviation(rsd)were 0.04%-1.08%,0.91%-5.36%respectively.The recovery rate of sample addition was more than 75%,and RSD ranged from 0.04%to 5.16%.Conclusion The method can be used for the determination of four chemical hypoglycemic agents.
