利用莱士曼氏乳酸杆菌检测婴幼儿配方乳粉中维生素B_(12)方法的建立与应用

摘要 本研究从自然发酵牛乳中分离鉴定出一株莱士曼氏乳酸杆菌,命名为LL-JG001株,基于LL-JG001株生长速率与维生素B_(12)含量的正相关性,绘制标准曲线,建立了检测婴幼儿配方乳粉中维生素B12含量的微生物法。结果显示,LL-JG001株生长速率与维生素B12含量线性关系良好,R^(2)为0.99;加标回收率为99.3%~101.2%,RSD<2%,符合加标回收要求;精密度试验中RSD值为2.6%,表明重复性较好。采集市售婴幼儿配方奶粉样本10份,利用建立的方法检测样本中维生素B_(12)含量,结果与商品化微生物法检测试剂盒结果相符,市售婴幼儿配方奶粉中维生素B_(12)含量达到国标要求。综上,本研究基于LL-JG001建立的婴幼儿配方乳粉中维生素B12含量的检测方法具有良好的检测准确性和稳定性,为保障奶制品安全提供了技术支持。 This study isolated and identified a strain of Lactobacillusleichmannii,named as LL-JG001,from naturally fermented milk.Based on the correlation between the growth rate of LL-JG001 and the content of Vitamin B_(12),a standard curve was established,and a microbiological method for the detection of vitamin B_(12) in infant formula powder was developed.2 The results showed a good linear relationship between the growth rate of strain JG001 and the Vitamin B_(12) content,with an R^(2) value of 0.99.The spike recovery rate was 99.3% to 101.2%,with an RSD of less than 2%,meeting the spike recovery requirements.The precision test showed an RSD value of 2.3%to 4.7%,indicating good repeatability.Ten commercial infant formula powder samples were tested using the established method,and the results were consistent with those obtained using commercial microbiological test kits,meeting the national standard requirements for the Vitamin B_(12) content in commercial infant formula powder.In conclusion,the method developed in this research for the evaluate of Vitamin B_(12) in infant formula powder based on LL-JG001 demonstrates good accuracy and stability,providing technical support for ensuring the safety of dairy products.
