摘要
阐明尿酸盐结晶(MSU)诱发慢性痛并转换为抑郁样行为的作用与机理。利用动物踝关节注射单钠尿酸盐结晶诱导急性痛风性关节炎,从而诱发小鼠抑郁样行为。行为学研究表明,注射MSU引起了持续性的踝关节疼痛,并显示显著的抑郁样行为,免疫荧光染色实验显示前额叶GABA神经元的显著改变,表明MSU诱发慢性痛和抑郁样行为。该抑郁模型具有造模时间短,造模后无肢体受损作用,不影响小鼠的活动能力,对相关行为学检测无不良影响,对于由慢性痛转换成抑郁症的深入研究具有实践意义。
To clarify the role and mechanism of monosodium urate(MSU)crystals in inducing chronic pain and converting it into depression-like behavior.Acute gouty arthritis was induced by injecting MSU crystals into the ankle joint of animals,thereby inducing depression-like behavior in mice.Behavioral studies showed that MSU caused persistent ankle pain and significant depression-like behavior.Immunofluorescence staining experiments revealed significant changes in GABA neurons in the prefrontal cortex,indicating that MSU induces chronic pain and depression-like behavior.This depression model has the advantages of short modeling time,no limb damage after modeling,no impact on the activity ability of mice,and no adverse effects on related behavioral tests,which is of practical significance for in-depth research on the conversion from chronic pain to depression.
