基于RhoA/ROCK通路研究复方中药对腹水肉鸡心肌损伤的作用

Effects of Compound Chinese Medicine on Improving Myocardial Damage in Ascites Broilers Based on RhoA/ROCK Pathway

摘要 为分析RhoA/ROCK信号通路在肉鸡腹水综合征(Ascites syndrome, AS)心肌损伤中的作用及复方中药的防治机制,本研究选取100羽健康的罗斯肉鸡(7日龄)并随机分成5组(每组20只),即基础饲粮饲养对照组,多因素法构建的AS模型组,复方中药高、低剂量治疗组以及L-精氨酸治疗组。观察临床症状和剖检变化,并通过HE染色和Masson染色分别观察35日龄肉鸡心脏组织结构变化和胶原纤维沉积;计算肉鸡腹水心脏指数(Ascites heart index, AHI);利用实时荧光定量PCR检测心脏组织MYPT1 mRNA的转录情况,ELISA法检测心脏组织RhoA/ROCK信号通路相关蛋白的表达。结果表明,与空白对照组相比,模型组肉鸡腹部膨大,触压有波动感,腹腔内呈现大量淡黄色液体,右心肥大,心脏组织结构排列紊乱,胶原纤维大量沉积,而且M组AHI显著升高(P <0.01);荧光定量PCR结果表明,心脏组织MYPT1 mRNA转录显著升高(P <0.01)。ELISA结果表明,RhoA、ROCK、P-MLC表达显著升高(P <0.01),而MLCP表达显著降低(P <0.01);与AS组相比,复方中药治疗组的肉鸡相关临床症状及上述指标明显改善。本研究发现,RhoA/ROCK通路介导的MLC磷酸化参与了AS中心肌损伤的发生,复方中药水提液可通过调节RhoA/ROCK抑制心肌损伤来有效防治AS。 The purpose of this study is to analyze the role of RhoA/ROCK pathway in myocardial injury of ascites syndrome(AS)and the prevention and treatment mechanism of compound Chinese medicine extract.One hundred 7-day-old Ross broilers were randomly divided into five groups.(20 per group).The control group was fed routinely,and the AS model group was built by low temperature,high energy diet and high salt drinking water.On the basis of the model group,the AS of broilers was treated by Chinese medicine extract with the high-dose and low-dose and L-Arg,respectively.The clinical symptoms and autopsy changes were observed.The heart tissue structure damage and collagen fiber deposition of 35-day-old broilers were evaluated by HE staining and Masson staining,respectively.The ascites heart index(AHI)of broilers was calculated.The relative expression of MYPT1 mRNA in heart tissues was analyzed by real-time fluorescence quanti-tative PCR,and the expression of RhoA/ROCK signal path related proteins in heart tissues were detected by ELISA.The results showed that compared with the control group,abdominal distension,fluctuating touch pressure,a large amount of yellowish liquid in abdominal cavity,hypertrophy of right heart,disordered arrangement of heart tissue structure and a large amount of collagen fiber deposition was observed in AS model group.AHI in M group increased significantly(P<0.01).The qPCR and the ELISA results showed that the MYPT1 mRNA tran-scription,and the expression of RhoA,ROCK and P-MLCP protein in heart tissue was significantly increased(P<0.01)and the activity of MLCP protein was significantly decreased.Compared with the AS group,the broilers in compound Chinese medicine group can improve the clinical symptoms and the above indexes.In conclusion,these results showed that MLC phosphorylation mediated by RhoA/ROCK pat-hway paly an important role in myocardial injury of ascites syndrome,and the extract of compound Chinese medicine can effectively prevent and treat AS,and the mechanism is closely related to the inhibition of myocardial injury by RhoA/ROCK pathway.
