摘要
为开发棘胸蛙(Quasipaa spinosa)蝌蚪的饲料,探究了不同饲料配方对棘胸蛙蝌蚪死亡率和变态率的影响及其分子机制.记录并分析了60 d处理后,在高蛋白添加复合维生素饲料(high-protein feed mixed with vitamins,HP_V)、高蛋白饲料(high-protein feed,HP)和低蛋白饲料(low-protein feed,LP)喂养下的棘胸蛙蝌蚪的变态率和死亡率,并对各组蝌蚪的肝脏组织进行转录组生物信息学分析.结果表明:HP_V组变态率显著高于HP组,HP组死亡率显著高于HP_V组和LP组;HP组和HP_V组的差异表达基因(differentially expressd genes,DEGs)在免疫通路富集,HP_V组的pIgR,TRIM25和DHX58等基因表达上调,HP组中MHCⅡ类基因家族、IL-17E和IL-17F等基因的表达上调,在HP_V和LP组中,DEGs在胆固醇合成通路富集,DHCR24等相关基因表达上调;在HP_V组中生长素抑制素(SST)基因表达下调,qPCR结果与转录组结果相一致.分析结果证实,高蛋白饲料饲养下的棘胸蛙蝌蚪可能会激活较强的免疫反应导致肝脏出现炎症,间接导致养殖过程中蝌蚪的死亡率上升,而在提高饲料蛋白质含量的同时适量补充维生素可以显著降低蝌蚪的死亡率,同时提高其变态率,这可能与HP组中胆固醇和甲状腺素的合成受到抑制等因素有关.
For the development of feed for Quasipaa spinosa tadpoles,it was investigated the impact of various feed formulations on tadpole mortality and metamorphosis rates and their molecular mechanisms.After 60-days of experimental treatment,it was observed and analyzed the metamorphosis and mortality rates of tadpoles fed with high-protein(HP),high-protein vitamin complex(HP_V),and low-protein(LP)diets.Additionally,transcriptome bioinformatics analysis was performed on liver tissues from each group.The findings revealed that the metamorphosis rate in the HP_V group was significantly higher than that in the HP group,while the mortality rate in the HP group was significantly higher than that in both the HP_V and LP groups.Differentially expressed genes(DEGs)associated with immune pathways were enriched in the HP and HP_V groups,with higher expression observed in genes such as pIgR,TRIM25,and DHX58 in the HP_V group.Conversely,the MHC II gene family,IL-17E,and IL-17F genes were upregulated in the HP group.Furthermore,DEGs related to cholesterol synthesis pathways exhibited upregulated expression of genes such as DHCR24 in the HP_V and LP groups.The expression of somatostatin(SST)was downregulated in the HP_V group,with qPCR results aligning with transcriptome findings.The analysis results indicated that feeding Q.spinosa tadpoles a high-protein diet might trigger a robust immune response,resulting in liver inflammation,and indirectly contributing to increase mortality during the breeding process.However,the addition of appropriate vitamin supplementation alongside increased protein content could significantly reduce mortality rates and improved the metamorphosis rate of Q.spinosa tadpoles,these effects was found to be associated with factors such as the inhibition of cholesterol and thyroxine synthesis in the HP group.
