不同分子修饰的普鲁士蓝纳米立方块的制备及对大肠杆菌的光热抗菌性能

Preparation of Prussian blue nanocubes modified with different molecules and their photothermal antimicrobial performance against Escherichia coli

摘要 以聚乙烯亚胺(polyethyleneimine,PEI)、透明质酸(hyaluronic acid,HA)和柠檬酸(citric acid,CA)为稳定剂,采用沉淀法分别制备了不同分子修饰的普鲁士蓝(Prussian blue,PB)纳米立方块(PB-x,x=PEI,HA,CA),同时在不使用稳定剂的条件下制备了纯普鲁士蓝纳米粒子.采用XRD,SEM,FT-IR,DLS表征产物的结构、形貌、化学组成和尺寸分布,考察了上述粒子的光学吸收能力、光热性质和光热抗菌性能,并对性能最优的PB-CA的光热转化效率、光热稳定性和储存稳定性进行了研究.结果表明:产物粒子在808 nm波长处的光吸收能力越强,光热性质就越出色,其中PB-CA的光热性质最佳,在4次升温-降温循环中表现出令人满意的光热稳定性;PB-CA对大肠杆菌的杀灭能力优于其他3种粒子,在0.8 W/cm^(2)的808 nm近红外光照射下10 min内可以完全杀灭细菌;PB-CA具有良好的储存稳定性.这些结果预示,PB-CA具有发展成为抗菌用新型光热剂的潜质. Prussian blue nanocubes(PB-x,x=PEI,HA,CA)modified with different molecules were prepared by precipitation method using polyethyleneimine(PEI),hyaluronic acid(HA)and citric acid(CA)as stabilizers.The pure Prussian blue nanoparticle(PB)was also prepared in the absence of any stabilizing agent.The structure,morphology,chemical composition and size distribution of the obtained products were characterized by XRD,SEM,FT-IR and DLS.Their optical absorption capacity,photothermal property and antimicrobial performance were tested,and the photo-thermal conversion efficiency,photothermal stability and storage stability of the optimized sample PB-CA were also investigated.The results showed that the particle with stronger absorption capacity at 808 nm wavelength exhibited more excellent photothermal property.Among these nanomaterials,PB-CA had the best photothermal property and demonstrated satisfactory photothermal stability after four heating-cooling cycles.The antimicrobial performance of PB-CA against Escherichia coli was superior to that of others.All the bacteria incubated with PB-CA were completely killed within 10 min under 808 nm near-infrared laser irradiation at a power density of 0.8 W/cm^(2).Furthermore,PB-CA showed good storage stability.All these results indicated that PB-CA had the potential to be developed into a new photothermal agent for antibacterial therapy.
