基于NF-κB信号通路研究桂枝茯苓丸抗小鼠肺癌血管生成的作用及相关因子

摘要 目的 基于NF-κB信号通路研究桂枝茯苓丸抗小鼠肺癌血管生成的作用及相关因子。方法 选取SPF级C57BL/6雌性小鼠80只,将其中60只随机分为模型组、沙利度胺组、低剂量组、中等剂量组、高剂量组及联合组,每组10只;将另外20只分为空白对照组、低中药组、中等中药组、高中药组,每组5只,测定相关指标。结果 灌胃第7、14、21天,沙利度胺组、低剂量组、中等剂量组、高剂量组、联合组的肿瘤体积小于模型组(P<0.05)。沙利度胺组、低剂量组、中等剂量组、高剂量组及联合组小鼠的瘤质量、微血管密度(MVD)及血管内皮生长因子(VEGF)、血管内皮生长因子受体(VEGFR)、Survivin蛋白免疫组化染色评分低于模型组(P<0.05)。低中药组、中等中药组、高中药组的VEGF、VEGFR、Survivin mRNA相对表达量低于空白对照组(P<0.05)。结论 桂枝茯苓丸可抑制NF-κB信号通路,调控肺癌细胞增殖、凋亡与基因表达,发挥抗肿瘤、抗血管生成作用。 Objective To investigate the mechanism of Guizhi Fuling pill in inhibiting angiogenesis of lung cancer in mouse and related factors based on the NF-κB signaling pathway.Methods A total of 80 SPF C57BL/6 female rats were selected,60 rats of them were randomly divided into model group,thalidomide group,low-dose group,medium-dose group,high-dose group and combined group,with 10 rats in each group;the other 20 rats were divided into blank control group,low Chinese medicine group,medium Chinese medicine group and high Chinese medicine group,with 5 rats in each group.The relevant parameters were measured.Results On the 7 th,14 th and 21 st day of gavage,the tumor volume of the thalidomide group,the low-dose group,the medium-dose group,the high-dose group and the combined group was smaller than that of the model group(P<0.05).The tumor weight,microvessel density(MVD)and the immunohistochemical staining scores of vascular endothelial growth factor(VEGF),vascular endothelial growth factor receptors(VEGFR)and Survivin in the thalidomide group,the low-dose group,the medium-dose group,the high-dose group and the combined group were lower than those in the model group(P<0.05).The relative expression levels of VEGF,VEGFR and Survivin mRNA in the low Chinese medicine group,the medium Chinese medicine group and the high Chinese medicine group were lower than those in the blank control group(P<0.05).Conclusion Guizhi Fuling pill can inhibit the NF-κB signaling pathway,regulate the proliferation,apoptosis and gene expression of lung cancer cells,and exert anti-tumor and anti-angiogenesis effects.
