雷公藤甲素对SLE患者PBMC中miR-7、PTEN及淋巴细胞比例的影响

摘要 目的探讨雷公藤甲素(TP)对系统性红斑狼疮(SLE)患者外周血单个核细胞(PBMC)中微小核糖核酸-7(miR-7)、磷酸酶和张力蛋白同源物(PTEN)及淋巴细胞比例的影响。方法本研究选取2022年9月至2023年8月医院收治的11例SLE患者作为SLE组,另选取11例同期健康体检者作为Control组。对比两组PBMC中B淋巴细胞标志蛋白(CD19)、T淋巴细胞标志蛋白(CD3)、自然杀伤(NK)细胞标志蛋白(CD56)的表达水平和细胞凋亡情况及miR-7、PTEN表达水平;采用不同浓度的TP对SLE组PBMC干预24 h,检测不同PBMC中miR-7、PTEN、CD3、CD19、CD56的表达水平。结果SLE组PBMC凋亡水平高于Control组(P<0.05);SLE组PBMC中miR-7表达水平高于Control组,PTEN蛋白表达水平低于Control组(P<0.05);SLE组PBMC中CD19表达水平高于Control组,CD3、CD56表达水平低于Control组(P<0.05);采用5、25 nM的TP干预后,miR-7、CD19的表达水平均低于干预前,PTEN信使核糖核酸(PTEN mRNA)、CD3、CD56的表达水平均高于干预前(P<0.05)。结论TP干预处理可降低SLE患者PBMC中miR-7、CD19的表达水平,增加PBMC中PTEN、CD3、CD56的表达水平。 Objective To investigate the effects of triptolide(TP)on the proportion of Microrna-7(miR-7),Phosphatase and tensin homologues(PTEN),and lymphocyte in peripheral blood mononuclear cells(PBMC)in patients with systemic lupus erythematosus(SLE).Methods In this study,11 SLE patients admitted to the hospital from September 2022 to August 2023 were selected as the SLE group,and 11 healthy subjects in the same period were selected as the Control group.The expression levels of B lymphocyte marker protein(CD19),T lymphocyte marker protein(CD3),natural killer(NK)cell marker protein(CD56)and cell apoptosis,as well as the expression levels of miR-7 and PTEN were compared between the two groups.PBMC in SLE group was treated with different concentrations of TP for 24 h,and the expression levels of miR-7,PTEN,CD3,CD19 and CD56 in different PBMC were detected.Results The apoptosis level of PBMC in SLE group was higher than that in Control group(P<0.05).The expression level of miR-7 in PBMC of SLE group was higher than that of Control group,and the expression level of PTEN protein was lower than that of Control group(P<0.05).The expression level of CD19 in PBMC of SLE group was higher than that of Control group,and the expression levels of CD3 and CD56 were lower than that of Control group(P<0.05).After intervention with 5 nM and 25 nM TP,the expression levels of miR-7 and CD19 were lower than before intervention,and the expression levels of PTEN messenger RNA(PTEN mRNA),CD3 and CD56 were higher than before intervention(P<0.05).Conclusion TP intervention can decrease the expression of miR-7 and CD19 in PBMC of SLE patients,and increase the expression of PTEN,CD3 and CD56 in PBMC.
