摘要
目的建立比格犬缺血性心肌病模型,比较钩藤不同有效部位对比格犬缺血性心肌病模型的治疗作用。方法雄性比格犬44只,分为假手术组(6只)、模型组(38只),通过分离并永久性结扎冠状动脉左前降支,诱导缺血性心肌病模型。于造模4周后选择36只模型动物,按EF随机分为模型对照组、消心痛组(0.8 mg/kg)、钩藤带钩茎醇提取物组(0.5 g药材/kg)、钩藤不带钩茎醇提取物组(0.5 g药材/kg)、钩藤带钩茎水提取物组(0.5g药材/kg)、钩藤不带钩茎水提取物组(0.5 g药材/kg),每组6只动物。各组动物于分组后进行给药,每天1次,连续给药8周。给药8周后检测各组动物心功能CO、EF、FS、LVEDV、LVESV变化,血清中BNP、COP、Ang-Ⅱ水平,心室肌组织TMEM16A蛋白表达量以及心脏组织形态的变化。结果模型比格犬CO、EF、FS均显著下降,LVEDV、LVESV显著升高,心脏组织可见心肌纤维片状坏死,间质大量炎症细胞浸润,病灶内脂肪组织浸润;钩藤带钩茎水提取物、钩藤带钩茎醇提取物均可显著改善比格犬心功能,可降低血清BNP、COP、Ang-II水平,显著减轻心脏病变程度;其中钩藤带钩茎水提取物对缺血性心肌病的治疗作用最佳,降低左心室肌TMEM16A蛋白表达。结论钩藤带钩茎水提取物对犬缺血性心肌病的治疗作用最佳,其作用机制可能与抑制TMEM16A的表达,降低Ang Ⅱ的水平有关。
Objective To establish a model of ischaemic cardiomyopathy in Beagle dogs and to compare the therapeutic effects of different effective parts of Uncaria rhynchophylla in a model of ischaemic cardiomyopathy in Beagle dogs.Methods 44 male Beagles,divided into a sham operation group(6 animals)and a model control group(38 animals),were used to induce an ischaemic cardiomyopathy model by isolating and permanently ligating the left anterior descending branch of the coronary artery.36 model animals were selected after 4 weeks of modelling and randomly divided by EF into the model control group,elimination of cardiac pain group(0.8 mg/kg),Alcohol Extract Group of Uncaria rhynchophylla Stem with Hooks(0.5 g herb/kg),Alcohol Extract Group of Uncaria rhynchophylla Stem without Hooks(0.5 g herb/kg),Water Extract Group of Uncaria rhynchophylla Stem with Hooks(0.5g herb/kg),Water Extract Group of Uncaria rhynchophylla Stem without Hooks(0.5g herb/kg),6 animals in each group.The animals in each group were administered once daily for 8 weeks after grouping.After 8 weeks of administration,the animals in each group were examined for changes in cardiac function CO,EF,FS,LVEDV,LVESV,serum levels of BNP,COP,Ang-ⅡTMEM16A protein expression in ventricular myocardial tissues as well as changes in the morphology of cardiac tissues.Results Model Beagle dogs CO,EF,FS were significantly decreased,LVEDV,LVESV significantly increased,cardiac tissue could be seen in cardiac myofibrillar sheet necrosis,a large number of interstitial inflammatory cells infiltration,foci of adipose tissue infiltration;Uncaria rhynchophylla with hooked stems aqueous extract,Uncaria rhynchophylla with hooked stems alcoholic extracts could significantly improve the cardiac function of the Beagle dogs,reduce the level of serum BNP,COP,Ang-Ⅱ,significantly reduce the degree of cardiac lesions;among them,the aqueous extract of Uncaria rhynchophylla with glochidia had the best therapeutic effect on ischemic cardiomyopathy and significantly increased the expression of left ventricular myocardial TMEM16A protein.Conclusion The aqueous extract of Uncaria rhynchophylla with hooked stems show the best therapeutic effect on canine ischaemic cardiomyopathy,and its mechanism of action may be related to the inhibition of the expression of TMEM16A and the reduction of AngIl levels.
