摘要
小麦条锈病是影响我国小麦生产的重要病害之一。为开发与高温成株抗条锈病基因Yr52连锁的分子标记。本研究以‘晋麦47’为感病亲本,‘PI 660057’(含有Yr52的单基因系)为抗病亲本构建遗传分离群体。结合F_(2)代单株及其对应的F_(2∶3)群体的抗、感病表型,选取F_(2∶3)群体内所有单株反应型均为2~3和8~9的F_(2)代单株各30株构建抗病池和感病池,并结合抗、感病亲本进行外显子捕获测序分析。在外显子捕获测序分析获得的目标区间内开发了9个KASP标记,对构建的另一个F′_(2)群体进行重组单株筛选和基因定位,在定位基础上又开发了3个KASP标记对F_(2∶4)群体进行重组单株筛选和基因定位。结果显示,高温成株期抗性基因Yr52定位于KASP-2123与KASP-2700两个标记之间,区间遗传距离为0.83 cM,对应于‘中国春’参考基因组(v 2.1)序列约570 kb的物理区间。本研究开发的Yr52基因两侧的连锁分子标记可用于分子标记辅助选择育种,也可用于Yr 52的精细定位与图位克隆。
Wheat stripe rust is a major disease affecting wheat production in China.This study aimed to develop molecular markers closely linked to the high-temperature adult-plant resistance gene Yr52.The susceptible parent‘JM47’and the resistant parent‘PI 660057’(a single-gene line containing Yr52)were used to construct a segregating population.Phenotyping was conducted on the F_(2)and F_(2∶3)populations.Leaf DNA from 30 plants in F_(2)population corresponding to F_(2∶3)plants with reaction types of 2-3 and 8-9 was used to construct resistance(R)and susceptible(S)bulks for bulk segregant exome capture sequencing(BSE-seq).Nine KASP markers were developed within the target interval obtained from BSE-seq analysis.An additional F′_(2)population was subjected to recombinant plant screening and gene fine mapping,leading to the development of three more KASP markers for the F_(2∶4)population.The results showed that the Yr52 was located between markers KASP-2123 and KASP-2700,with a genetic distance of 0.83 cM,corresponding to a physical interval of approximately 570 kb in the‘Chinese Spring’reference genome(v 2.1).The molecular markers developed in the study,closely linked to Yr52,can be applied in marker-assisted selection for the stripe rust resistance breeding and used for fine mapping and gene cloning of Yr52.
