摘要
为明确球孢白僵菌Beauveria bassiana与鱼藤酮联用对草地螟Loxostege sticticalis幼虫的防治效果,本研究通过室内生物测定比较6株球孢白僵菌对草地螟2龄幼虫的致病力,筛选获得高致病力菌株;利用该高致病力菌株与植物源农药鱼藤酮联用防治草地螟幼虫,以获得联用效果;通过比较球孢白僵菌、鱼藤酮联用处理相对于单剂处理下草地螟的防御酶活性变化,探讨菌药增效机制。结果表明,6株球孢白僵菌中JDB菌株对幼虫的致病力最强,用孢子浓度1×10^(8)个/mL的悬浮液处理草地螟幼虫10 d后,校正死亡率达88.53%。将3个浓度的JDB孢子悬浮液(1×10^(8)、1×10^(7)个/mL和1×10^(6)个/mL)分别与LC_(10)(1.60 mg/L)和LC_(20)(2.95 mg/L)鱼藤酮混配,其中,1×10^(7)个/mL+LC_(20)、1×10^(7)个/mL+LC_(10)、1×10^(6)个/mL+LC_(20)和1×10^(6)个/mL+LC_(10)4个组合在第10天均表现为增效作用,χ^(2)均大于3.841,分别为8.76、6.07、14.99和13.79。酶活性测定结果表明,球孢白僵菌与鱼藤酮联用处理的试虫体内超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性在处理后48 h和60 h显著高于各单剂处理,羧酸酯酶(CarE)的活性在处理后60 h显著高于各单剂处理。综上所述,鱼藤酮对球孢白僵菌JDB防治草地螟具有协同增效作用。
The pathogenicity of six Beauveria bassiana strains against larvae of Loxostege sticticalis was tested in the laboratory to select the most virulent strain for combined use with the plant-derived pesticide rotenone.The synergistic mechanism of fungal and pesticide treatment was explored by comparing the changes in defense enzyme activities of L.sticticalis under combined treatment compared to single-agent treatment.The results showed that the JDB strain was the most virulent among the tested B.bassiana strains,achieving a corrected mortality rate of 88.53%after 10 days when larvae were treated with a suspension containing 1×10^(8) spores/mL.Three concentrations of JDB spore suspensions(1×10^(8),1×10^(7)and 1×10^(6)spores/mL)were combined with two doses of rotenone(LC_(20):2.95 mg/L and LC_(10):1.60 mg/L).Synergistic effects were observed on the 10th day for combinations of 1×10^(7)spores/mL+LC_(20),1×10^(7)spores/mL+LC 10,1×10^(6)spores/mL+LC_(20)and 1×10^(6)spores/mL+LC_(10),withχ^(2)values exceeding 3.841(8.76,6.07,14.99 and 13.79,respectively).Enzyme activity analysis revealed that the combined treatment significantly increased superoxide dismutase(SOD)and catalase(CAT)activities at 48 h and 60 h compared to single-agent treatments.Additionally,the activity of carboxylesterase(CarE)was significantly higher than that in single-agent treatments at 60 h.In conclusion,the combination of rotenone with the JDB strain of B.bassiana demonstrated a synergistic effect,enhancing its virulence against L.sticticalis larvae.
