摘要
目的:运用高分辨质谱技术分析鉴定拯阳煎的化学成分及小鼠灌胃后的吸收成分。方法:将健康雄性小鼠12只分为空白组3只和给药组9只。空白组灌胃超纯水;给药组按30 mL/kg灌胃拯阳煎提取液,收集血清和组织,采用蛋白沉淀的方法进行样品前处理。质谱采用正负离子一级全扫描/数据依赖二级扫描模式采集数据,利用提供的离子碎片相对保留时间、二级碎片信息,经Xcalibur 4.2软件拟合元素组成,再通过对照品、文献以及数据库信息比对,实现对拯阳煎化学成分及入血、组织原型成分的鉴定。结果:共鉴定263种化学成分,包括黄酮类99种、皂苷类37种、木脂素类21种、生物碱类20种、有机酸类19种、萜类16种、苯酞类12种、香豆素类7种、苯乙醇苷类5种、苯丙素类4种以及其他类23种。小鼠灌胃拯阳煎后分别于0.5 h、1 h、3 h时取样检测,检测血清、肺脏、心脏、肝脏、脾脏、肾脏、脑组织中原型成分。血清中分别检测到167、167、91种;肺中140、123、95种;心脏中94、105、81种;肝脏中137、150、85种;脾脏中138、14、13种;肾脏中171、177、158种;脑组织中51、59、43种。结论:利用高分辨质谱技术完成了拯阳煎化学成分及主要入血、组织移行成分的快速鉴定,可为进一步开展拯阳煎效应物质的筛选及入血成分体内药代动力学研究提供参考。
Objective:To analyze and identify the chemical constituents of Zhengyang Decoction and the absorbed components in mice after oral administration using high-resolution mass spectrometry.Methods:Twelve healthy male mice were divided into a blank group(n=3) and a treatment group(n=9).The blank group received ultrapure water via gavage,while the treatment group was administered Zhengyang Decoction extract at a dose of 30 mL/kg.Serum and tissue samples were collected and pretreated using protein precipitation.Mass spectrometry data were acquired in both positive and negative ion modes using full MS1 scan and data-dependent MS2 scan modes.Using the provided ion fragments,relative retention time,and secondary fragmentation information,the elemental compositions were fitted using Xcalibur 4.2 software.Subsequently,by comparing with reference standards,literature,and database information,the chemical constituents of Zhengyang Decoction,as well as its prototype components found in blood and tissues,were identified.Results:A total of 263 chemical constituents were identified,including 99 flavonoids,37 saponins,21 lignans,20 alkaloids,19 organic acids,16 terpenes,12 phthalides,7 coumarins,5 phenylethanoid glycosides,4 phenylpropanoids,and 23 other compounds.After oral administration of Zhengyang Decoction,samples were collected at 0.5 h,1 h,and 3 h,and prototype compounds were detected in serum and various tissues including lung,heart,liver,spleen,kidney,and brain.The number of prototype compounds detected were as follows.Serum:167,167,91;Lung:140,123,95;Heart:94,105,81;Liver:137,150,85;Spleen:138,14,13;Kidney:171,177,158;Brain:51,59,43.Conclusion:High-resolution mass spectrometry enabled rapid identification of the chemical constituents of Zhengyang Decoction and its major blood-absorbed and tissue-distributed prototype compounds.These findings provide a foundation for further screening of active substances and pharmacokinetic studies of blood-absorbed components of Zhengyang Decoction.
