摘要
目的观察黄精多糖(PSP)对高血压左室肥厚伴心力衰竭大鼠血压及心脏功能的影响。方法建立大鼠高血压左室肥厚伴心力衰竭模型24只,随机分为模型组、PSP低剂量组、PSP中剂量组、PSP高剂量组,每组6只,另选择同源Wistar大鼠6只为对照组。PSP低剂量组、PSP中剂量组、PSP高剂量组分别给予PSP水溶液500、800、1000 mg·kg^(-1)·d^(-1)灌胃,模型组和对照组给予等量生理盐水灌胃。连续灌胃8周后,比较各组大鼠血压、心脏功能指标[左室收缩期前壁厚度(LVAWS)、左室舒张期前壁厚度(LVAWD)、左室收缩期后壁厚度(LVPWS)、左室舒张期后壁厚度(LVPWD)、左室收缩末期内径(LVESD)、左室舒张末期内径(LVEDD)、左室射血分数(LVEF)和左室短轴缩短率(LVFS)]及心脏组织改变。结果建模后,模型组尾动脉收缩压(SBP)高于对照组(P<0.05);灌胃4周、8周后,PSP中剂量组、PSP高剂量组尾动脉SBP较模型组降低(P<0.05),而PSP低剂量组与模型组比较差异无统计学意义(P>0.05)。灌胃8周后,模型组LVAWS小于对照组(P<0.05),PSP中剂量组、PSP高剂量组LVAWS较模型组升高(P<0.05),而PSP低剂量组与模型组比较差异无统计学意义(P>0.05);模型组LVESD小于对照组,LVEF及LVFS低于对照组(P<0.05),PSP低剂量组、PSP中剂量组LVESD较模型组增大,PSP低剂量组、PSP中剂量组及PSP高剂量组LVEF和LVFS较模型组升高(P<0.05);5组LVAWD、LVPWS、LVPWD及LVEDD比较差异均无统计学意义(P>0.05)。结论PSP能改善高血压左室肥厚伴心力衰竭大鼠血压及心脏功能,具体机制有待进一步研究。
Objective To observe the effect of polygonatum sibiricum polysaccharides(PSP)on blood pressure and cardiac function in hypertensive rats with left ventricular hypertrophy and heart failure.Methods Twenty-four rats were es tablished as hypertensive left ventricular hypertrophy with heart failure model,and randomly divided into model group,PSP low-dose group,PSP medium-dose group,PSP high-dose group,6 rats in each group,and 6 homozygous Wistar rats were se lected as control group.The PSP low-dose group,PSP medium-dose group and PSP high-dose group were gavaged with 500,800 and 1000 mg·kg^(-1)·d^(-1)of aqueous PSP solution,respectively,and the model group and control group were ga vaged with equal amounts of saline.After 8 weeks of continuous gavage,the blood pressure,cardiac function and cardiac tissue changes were compared among the groups.Results After modeling,the caudal artery systolic blood pressure(SBP)of the model group was higher than that of the control group(P<0.05).After 4 and 8 weeks of gavage,the SBP of the PSP medium-dose group and PSP high-dose group were lower than that of the model group(P<0.05),while the difference between the PSP low-dose group and the model group was not statistically significant(P>0.05).After 8 weeks of gavage,LVAWS was reduced in the model group compared with the control group(P<0.05),LVAWS was elevated in the PSP medium-dose group and PSP high-dose group compared with the model group(P<0.05),while the difference between the PSP low-dose group and the model group was not statistically significant(P>0.05).The LVESD,LVEF and LVFS of the model group were lower than those of the control group(P<0.05).LVESD widened in the PSP low-dose group and PSP medium-dose group compared with the model group,and LVEF and LVFS were elevated in the PSP low-dose group,PSP medium-dose group and PSP high-dose group compared with the model group(P<0.05).There were no statistically significant differences in the five groups for LVAWD,LVPWS,LVPWD and LVEDD(P>0.05).Conclusion PSP can improve blood pressure and cardiac function in hypertensive left ventricular hypertrophy with heart failure rats,and the specific mechanism needs further research.
