摘要
目的分析丹参酮ⅡA对原发性痛经模型大鼠的治疗效果及其相关机制。方法将60只SD大鼠,按体质量随机分为丹参酮ⅡA(10 mg/kg)组、丹参酮ⅡA(30 mg/kg)组、丹参酮ⅡA(60 mg/kg)组,吲哚美辛(6.75 mg/kg)组,模型组和空白对照组,每组10只。除空白对照组外,其余各组大鼠于腹股沟区域皮下注射苯甲酸雌二醇注射液结束间隔24 h后,腹腔注射缩宫素注射液制备模型。丹参酮ⅡA各组大鼠于造模第5天开始灌胃给药,吲哚美辛组于造模第9天开始灌胃给药,空白对照组仅给予等体积蒸馏水。记录各组大鼠30 min内扭体次数及疼痛潜伏期时间;炎性指标[白介素(IL)-1、IL-6]、氧化应激指标[超氧化物歧化酶(SOD)、丙二醛(MDA)]、疼痛因子[前列腺素E_(2)(PGE_(2))、前列腺素F_(2α)(PGF_(2α))]。结果与空白对照组比较,模型组扭体次数增加、扭体潜伏期缩短(P<0.01);与模型组比较,丹参酮ⅡA(60mg/kg)组及吲哚美辛(6.75mg/kg)组扭体次数减少,疼痛潜伏期延长(P<0.05或P<0.01),丹参酮ⅡA(30mg/kg)组疼痛潜伏期延长(P<0.01)。与空白对照组比较,模型组血清IL-1、IL-6水平均升高(P<0.01);与模型组比较,丹参酮ⅡA(60 mg/kg)组、丹参酮ⅡA(30 mg/kg)组及吲哚美辛(6.75 mg/kg)组血清IL-1、IL-6水平均降低(P<0.01或P<0.05)。与空白对照组比较,模型组血清MDA水平升高,血清SOD水平降低(P<0.01);与模型组比较,丹参酮ⅡA(60 mg/kg)组、丹参酮ⅡA(30 mg/kg)组及吲哚美辛(6.75 mg/kg)组血清MDA水平降低,SOD水平升高(P<0.01)。与空白对照组比较,模型组子宫组织PGF_(2α)水平升高,PGE_(2)含量降低(P<0.01);与模型组比较,丹参酮ⅡA(60 mg/kg)组、丹参酮ⅡA(30 mg/kg)组及吲哚美辛(6.75 mg/kg)组子宫组织PGF_(2α)含量均降低,PGE_(2)含量升高(P<0.05或P<0.01)。结论高剂量丹参酮ⅡA可通过改善痛经大鼠的氧化应激反应、疼痛指标,抑制子宫炎性因子产生等机制治疗原发性痛经。
Objective To analyze the therapeutic effect of tanshinoneⅡA on primary dysmenorrhea in rats and its related mechanism.Methods Sixty SD rats were randomly divided into tanshinoneⅡA(10 mg/kg)group,tanshinoneⅡA(30 mg/kg)group,tanshinoneⅡA(60 mg/kg)group,indomethacin(6.75 mg/kg)group,model group and blank control group,with 10 rats in each group.Except for the blank control group,the rats in the other groups were injected subcutaneously with estradiol benzoate injection in the inguinal region at an interval of 24 h,and the model was prepared by intraperitoneal injection of oxytocin injection.The rats in the tanshinoneⅡA group were given intragastric administration on the 5th day of modeling,the indomethacin group was given intragastric administration on the 9th day of modeling,and the blank control group was only given the same volume of distilled water.The number of twists and pain latency within 30 min and inflammatory factors(IL-1,IL-6),oxidative stress indexes(SOD,MDA)pain factors(PGE_(2),PGF_(2α))were recorded.Results Compared with the blank control group,the number of twists in the model group increased and the incubation period of twists was shortened(P<0.01).Compared with the model group,the number of twists and the pain latency were prolonged in the tanshinoneⅡA(60 mg/kg)group and the indomethacin 6.75 mg/kg group(P<0.05 or P<0.01),and the pain latency was prolonged in the tanshinoneⅡA(30 mg/kg)group(P<0.01).Compared with the blank control group,the serum levels of IL-1 and IL-6 in the model group increased(P<0.01).Compared with the model group,the serum levels of IL-1 and IL-6 in the tanshinoneⅡA(60 mg/kg)group,tanshinoneⅡA(30 mg/kg)group and indomethacin(6.75 mg/kg)group were decreased(P<0.01 or P<0.05).Compared with the blank control group,the serum MDA level and the serum SOD level decreased in the model group(P<0.01).Compared with the model group,the serum MDA level increased,and the serum SOD level decreased in tanshinone II.A(60 mg/kg),tanshinoneⅡA(30 mg/kg)group and indomethacin(6.75 mg/kg)group(P<0.01)Compared with the blank control group,the level of PGF_(2α)in uterine tissue in the model group increased,and the content of PGE_(2)decreased(P<0.01).Compared with the model group,the content of PGF_(2α)in uterine tissues decreased in tanshinoneⅡA(60 mg/kg),tanshinoneⅡA(30 mg/kg)group and indomethacin(6.75 mg/kg)group,and PGE_(2)content increased(P<0.05 or P<0.01).Conclusion TanshinoneⅡA can treat primary dysmenorrhea by improving oxidative stress response,pain indexes,and inhibiting the production of uterine inflammatory factors in rats with dysmenorrhea.
