桶状海绵共生真菌Penicillium rolfsii 8196#次级代谢产物

The secondary metabolites of the marine-derived fungus Penicillium rolfsii 8196 isolate from sponge Xestospongia sp.

摘要 对桶状海绵共生真菌Penicillium rolfsii 8196#的次级代谢产物进行了研究,通过大米固体发酵培养、多种色谱技术分离纯化以及核磁共振、质谱等波谱数据分析,鉴定了9个化合物3,5-dichloro-2,6-dihydroxy-4-methyl-benzoic acid methyl ester(1)、methyl asterrate(2)、penicillither(3)、methyl dichloroasterrate(4)、2,4-dichloroasterric acid(5)、asterric acid(6)、5-hydroxymethylasterric acid(7)、sulochrin(8)、hydroxysulochrin(9),其中化合物1、2、7首次从青霉属真菌中分离得到。MTT法测试细胞毒活性,表明化合物6对T-24细胞有细胞毒性,IC_(50)值为16.60μmol/L;二倍稀释法抗菌活性测试显示,化合物4、5对沙门氏菌有弱抗菌活性,其MIC值均为50μmol/L;ABTS自由基清除实验中发现化合物8和9在浓度为10μmol/L时具有强抗氧化活性,自由基清除率分别达到了79.81%、70.84%。 Nine known aromatic compounds(1-9)were isolated from the ethyl acetate extracts of the solid cultures of marie-derived fungus Penicillium rolfsii 8196#,isolated from sponge xestospongia sp..Their structures were elucidated by spectroscopic analysis and comparative investigation with references.These compounds were evaluated for their cytotoxic,antibacterial and antioxidant activities.Among them,Compound 6 showed cytotoxicity against human bladder cancer T-24 cells with the IC_(50) values of 16.60μmol/L,compounds 4 and 5 exhibited weak inhibitory effects against Salmonella with MIC values of 50μmol/L,and compounds 8 and 9 showed the free radical scavenging rate 79.81%and 70.84%at the concentration of 10μmol/L,respectively.
