谷子花青苷合成相关基因的鉴定与生物信息学分析

Identification and Bioinformatic Analysis of Genes Related to Anthocyanin Biosynthesis in Setaria italica

摘要 花青苷在植物生长发育过程中具有多种生理功能,鉴定谷子花青苷合成相关基因,可为谷子花青苷合成机制与相关基因功能研究提供理论依据。利用谷子的全基因组数据库以及水稻和拟南芥花青苷合成相关基因,通过多种生物信息软件及在线网站对谷子花青苷合成相关基因进行鉴定,并分析其蛋白质理化性质、系统进化、基因位置、基因结构、启动子顺式作用元件、GO注释与组织表达情况。在谷子中共比对到20类共59个花青苷合成相关基因;谷子与水稻花青苷合成相关基因有更近的亲缘关系;同一类谷子花青苷合成相关基因的基因结构较为相似;谷子花青苷合成相关基因的启动子顺式作用元件中,与光、茉莉酸甲酯和脱落酸相关的响应元件个数最多;谷子花青苷合成相关基因的表达具有组织特异性,大部分基因的表达在灌浆期达到最高水平,在成熟籽粒中只有个别基因高表达。谷子大部分花青苷合成相关基因的表达在灌浆期达到最高水平,光、干旱胁迫、茉莉酸甲酯和脱落酸与谷子花青苷合成存在紧密联系。 Anthocyanin has many physiological functions in plant growth and development.Identification of genes related to anthocyanin biosynthesis to provide theoretical basis for the study of anthocyanin biosynthesis mechanism and related gene functions in foxtail millet.In this study,genes related to anthocyanin biosynthesis in foxtail millet had been identified through the whole-genome database of foxtail millet and genes related to anthocyanin biosynthesis in rice and Arabidopsis.Then,the physical and chemical properties of proteins,phylogenetic relationship,gene location,gene structure,promoter cis-acting elements,tissue expression and GO annotation of genes related to anthocyanin biosynthesis in foxtail millet were analyzed through bioinformatics software and online websites.A total of 59 genes related to anthocyanin biosynthesis in 20 categories were identified in foxtail millet.Genes related to anthocyanin biosynthesis in the same category have the relatively similar gene structures in foxtail millet and rice.The gene structures of genes related to anthocyanin biosynthesis in the same type of foxtail millet are relatively similar.Among the promoter cis-acting elements of genes related to anthocyanin biosynthesis in Setaria italica,the largest number of response elements were associated with light,methyl jasmonate and abscisic acid.The expression of genes related to anthocyanin biosynthesis was tissue-specific,and most of the genes had the highest at filling stage,but only a few genes were highly expressed in mature seeds.Most genes related to an thocyanin biosynthesis in foxtail millet had the highest expression level during the filling stage.Light,drought stress,methyl jasmonate and abscisic acid were closely related to anthocyanin biosynthesis in foxtail millet.
