摘要
大熊猫栖息地土壤含有丰富的微生物资源,这些微生物不仅是大熊猫共生微生物的直接来源,还可通过影响土壤理化性质和主食竹的生长发育来间接影响大熊猫的健康。因此,以陕西佛坪国家级自然保护区内的大熊猫核心分布区内巴山木竹Bashania fargesii林为研究地点,采集冬春两季的土壤样品,通过细菌16S rRNA和真菌ITS基因测序,解析栖息地土壤微生物群落组成和多样性,以及群落内微生物间的互作关系。结果表明:(1)土壤中的细菌以变形菌门Proteobacteria、酸杆菌门Acidobacteria和放线菌门Actinobacteria为主;真菌则主要为子囊菌门Ascomycota、担子菌门Basidiomycota和罗兹菌门Rozellomycota;(2)细菌和真菌的α多样性均为冬季显著大于春季(P<0.05),β多样性无显著差异(P>0.05);(3)共现网络分析结果显示,微生物群落间的互作关系均以正相关关系为主,共鉴定出21个关键菌群。研究结果表明大熊猫栖息地土壤中微生物组成和多样性存在季节性差异,同时揭示微生物间的互作信息,为大熊猫栖息地微生态系统功能的提升提供了科学依据。
The soil in the giant panda habitat is rich in microbial source.The microorganisms are not only the direct sources of symbiotic microorganisms of giant pandas,but also indirectly affect the health of giant pandas by affecting the physicochemical properties of soil and the growth and development of dietary bamboos.Therefore,soil samples from the core habitat of giant panda in the Foping National Nature Reserve were collected from winter to spring in the wood bamboo(Bashania fargesii)forest,and the composition and diversity of soil microbial communities and the interactions among microbial communities were analyzed by sequencing the 16S rRNA of bacteria and ITS gene of fungi.The results showed that:(1)the main bacterial compositions in soil were Proteobacteria,Acidobacteria and Actinobacteria,and the main fungi were Ascomycota,Basidiomycota and Rozellomycota;(2)the α-diversity of both bacteria and fungi were significantly greater in winter than those in spring(P<0.05),while no significant difference was detected inβ-diversity(P>0.05);(3)the result of co-occurrence network analysis demonstrated the positive correlation between microbial communities in the whole soil,and a total of 21 key flora were identified.Moreover,there were markedly seasonal differences in soil microbial composition and diversity,and thus provides a scientific basis for the improvement of microbial ecosystem function in the giant panda’s habitat.
