紫苏种子中α-亚麻酸积累关键基因的筛选及克隆

Screening and cloning of key genes forα-linolenic acid accumulation in Perilla Frutescens seeds

摘要 为深入研究α-亚麻酸积累过程中的关键基因,以高α-亚麻酸含量紫苏品种43号为研究对象,选取油脂合成相关的6个关键酶基因,以茎为对照,利用qRT-PCR技术筛选在种子不同发育时期的表达量显著增高的关键基因。对筛选的关键基因进行生物信息学分析,并在紫苏中进行克隆。研究结果明确了候选基因PfFAD 3和PfFAD7基因是参与紫苏种子中α-亚麻酸积累的关键基因,并解析了PfFAD3和PfFAD7氨基酸及蛋白结构及与亲缘物种间的进化关系,为后续研究奠定了一定的基础。 To investigate the key genes involved in the accumulation ofα-linolenic acid,this paper chose No.43,a highα-linolenic acid content variety as the research object with Six key enzyme genes related to oil synthesis selected,the stem as the control,qRT-PCR technology as the tool to screening the significantly increased expression levels of key genes at different stages of seed development.Bioinformatics analysis and gene clone were performed in Perilla frutescens.The results indicated that candidate genes PfFAD3 and PfFAD7 were the key genes involved in the accumulation ofα-linolenic acid in Perilla Frutescens seeds.Moreover,it explored the amino acid and protein structures of PfFAD3 and PfFAD7,as well as their evolutionary relationships with related species,which may laya certain foundation for future research.
