摘要
目的研究南海来源真菌Aspergillus sp.BBW次级代谢产物及抗弧菌活性。方法通过凝胶柱层析、半制备高效液相等色谱技术对次级代谢产物进行分离和纯化,借助核磁共振等波谱技术结合文献鉴定化合物结构,利用牛津杯法测定化合物的抗菌活性。结果从海洋真菌Aspergillus sp.BBW中分离鉴定了8个化合物,分别为phomaligol A(1)、dihydropenicillic acid(2)、penicillic acid(3)、5-hydroxy-3-isopropyl-4-methoxyfuranone(4)、4-methyl-3-carboxylpyrazole(5)、3,4-dihydroxyphenylacetic acid methyl ester(6)、4-hydroxyphenylacetic acid methyl ester(7)、2-hydroxyphenylacetic acid methyl ester(8);化合物1、4、5、6、7、8对副溶血性弧菌(Vibrio parahemolyticus)的最低抑菌浓度(MIC)为3.15~19.09 mg/mL;化合物5、7、8对哈维氏弧菌(Vibrio harveyi)的MIC为3.15~9.65 mg/mL。结论海洋真菌Aspergillus sp.BBW次级代谢产物抗弧菌活性较好,具有抗弧菌开发潜力。
Objective To study secondary metabolites and anti-vibrio activity of the fungus Aspergillus sp.BBW collected from the South China Sea.Methods Secondary metabolites were separated and purified by chromatographic techniques(Sephadex LH-20 and semi-preparative High Performance Liquid Chromatography).The structures of the compounds were identified by nuclear magnetic resonance spectrum and compared with reported data.The anti-vibrio activity of the compounds was determined by the Oxford cup method.Results Eight compounds were isolated and identified as phomaligol A(1),dihydropenicillic acid(2),penicillic acid(3),5-hydroxy-3-isopropyl-4-methoxyfuranone(4),4-methyl-3-carboxylpyrazole(5),3,4-dihydroxyphenylacetic acid methyl ester(6),4-hydroxyphenylacetic acid methyl ester(7),2-hydroxyphenylacetic acid methyl ester(8).The minimum inhibitory concentration(MIC)of compounds 1,4,5,6,7,and 8 for Vibrio parahaemolyticus was 3.15−19.09 mg/mL.The MIC of compounds 5,7,and 8 for Vibrio harveyi was 3.15−9.65 mg/mL.Conclusion The good anti-vibrio activity of secondary metabolites separated from the marine fungus Aspergillus sp.BBW,has good potential for developing anti-vibrio.
