铜基金属有机框架改性聚醚醚酮体外抗菌及成骨性能研究

In Vitro Antibacterial and Osteogenic Properties of Copper-Based Metal-Organic Framework Modified Polyether Ether Ketone

摘要 目的 采用铜基金属有机框架(Cu-MOFs)改性聚醚醚酮(PEEK),体外评价其抗菌和成骨性能。方法 采用水浴法合成Cu-MOFs并对其进行形貌、粒径等表征。采用磺化处理-原位生长法将Cu-MOFs负载于PEEK表面,扫描电镜(SEM)观察其表面形貌,分别采用原子力显微镜(AFM)和水接触角测量评估表面粗糙度及亲水性,电感耦合等离子质谱法(ICP-MS)检测铜离子的释放。采用细菌死活染色、平板计数法评估改性材料抗菌性能,碱性磷酸酶染色和茜素红S染色法研究改性材料促成骨分化性能,细胞活力检测试剂盒(CCK-8)和细胞活死染色技术评价改性材料的细胞相容性。结果 成功制备平均粒径约150 nm的Cu-MOFs粒子,并采用磺化处理-原位生长法将Cu-MOFs负载于PEEK表面,制备了铜基金属有机框架改性的聚醚醚酮(sPEEK/MOFs),sPEEK/MOFs表面粗糙度是未改性材料的1.94倍,水接触角降低了45.8°,Cu ^(2+)在28 d累计释放量约为10.6 mg/L。sPEEK/MOFs对大肠杆菌和金黄色葡萄球菌的抗菌率分别达到89.3%和86.5%,明显高于未改性材料(P<0.001);sPEEK/MOFs能够提高MC3T3-E1细胞的碱性磷酸酶表达并能明显促进细胞钙化结节形成。结论 本研究制备的sPEEK/MOFs具有较好的抗菌和促成骨分化能力。 Objective To modify polyether ether ketone(PEEK)with copper-based metal-organic frameworks(Cu-MOFs)and evaluate its antibacterial and osteogenic properties in vitro.Methods Firstly,Cu-MOFs nanoparticles were synthesized via the water bath method,with their morphology and size characterized.Then,the sulfonation-in-situ growth method was used to load Cu-MOFs onto PEEK surfaces.Surface morphology was observed via scanning electron microscopy(SEM),surface roughness and wettability were assessed by atomic force microscopy(AFM)and contact angle measurements,and copper ion release was detected by inductively coupled plasma mass spectrometry(ICP-MS).Finally,the antibacterial properties of the modified PEEK were evaluated using live/dead bacterial staining and the plate technique,while the osteogenic differentiation-promoting ability was studied via alkaline phosphatase staining and alizarin red S staining.The cytocompatibility of the modified material was evaluated in vitro using cell counting kit(CCK-8)and live/dead cell staining.Results Cu-MOFs nanoparticles with an average particle size of about 150 nm were successfully prepared.The in-situ growth method successfully loaded Cu-MOFs onto the surface of PEEK,producing a copper-based metal-organic framework-modified PEEK(sPEEK/MOFs).The surface roughness of sPEEK/MOFs was 1.94 times that of the unmodified material,the water contact angle was reduced by 45.8°,and the cumulative release of Cu ^(2+)over 28 d was approximately 10.6 mg/L.In vitro antibacterial experiments showed that sPEEK/MOFs had antibacterial rates of 89.3%against Escherichia(E.coli)and 86.5%against Staphylococcus aureus(S.aureus),significantly higher than those of the unmodified material(P<0.001).In vitro osteogenic ability tests indicated that sPEEK/MOFs significantly increased the alkaline phosphatase expression of MC3T3-E1 cells and markedly promoted the formation of cellular calcified nodules.Conclusion sPEEK/MOFs possesses good antibacterial and osteogenic differentiation capabilities.
