摘要
【目的】PLAC8家族蛋白在植物调控器官大小及重金属转运方面具有重要作用,对该家族的研究有助于提高油茶的产量及品质。【方法】基于油茶基因组、转录组及单细胞转录组数据,利用生物信息学的方法对其PLAC8家族蛋白进行鉴定与分析。【结果】油茶基因组中含有26个CoPLAC8基因;系统进化树分析表明,CoPLAC8可以分为3个亚家族、7个分支,其中,SlFW2.2、MdCNR8、PaCNR12等3个与器官大小相关的基因分别分布在I-2、I-4、I-5分支,CoPLAC8-5与MdCNR8在同一亚支;Zn载体AtPLAC86分布在I-1分支,参与Cd转运的AtPLAC88、AtPLAC85及AtPLAC812分别分布在I-2、I-3、I-4分支。基因表达分析表明,CoPLAC8-5、CoPLAC8-7、CoPLAC8-17、CoPLAC8-18、CoPLAC8-20、CoPLAC8-23等6个基因在种子、种皮、果皮、茎、根等组织中的表达水平较高,CoPLAC8-5及CoPLAC8-18在成熟子房的心皮内层及胎座的表达水平较低。【结论】CoPLAC8-5可能是参与油茶果实大小调控的关键基因,CoPLAC8-5、CoPLAC8-7、CoPLAC8-17、CoPLAC8-18、CoPLAC8-20、CoPLAC8-23等6个基因可能与种仁中的Cu富集有关。
【Objective】PLAC8(Placenta-specific 8)family proteins play vital roles in regulating plant organ size and transporting heavy metals.Investigation of PLAC8s will help to promote both yield and quality in Camellia oleifera.【Method】Based on the genome,transcriptome and single-cell RNA-seq data,identification and analysis of PLAC8s were conducted in C.oleifera using bioinformatic approaches.【Result】A total of 26 CoPLAC8 genes were identified in the genome data of C.oleifera,and the corresponding proteins were divided into 3 subfamilies and 7 groups according to the topology of phylogenetic tree.SlFW2.2,MdCNR8 and PaCNR12,which were related with organ size,were distributed in group I-2,I-4,and I-5,respectively.CoPLAC8-5 was in the same subgroup with MdCNR8.AtPLAC86,that was a Zn exporter,was distributed in group I-1,while AtPLAC88,AtPLAC85 and AtPLAC812 which participated in Cd transport,were distributed in group I-2,I-3,and I-4,respectively.The results of transcriptome expression profiling showed that the expression of CoPLAC8-5,CoPLAC8-7,CoPLAC8-17,CoPLAC8-18,CoPLAC8-20 and CoPLAC8-23 were higher in seed,seed coat,pericarp,stem,and root.Single-cell RNA-seq revealed that the expression of both CoPLAC8-5 and CoPLAC8-18 were lower in inner layer of ovary wall and placenta of mature ovaries.【Conclusion】CoPLAC8-5 may be a key gene to regulate fruit size in C.oleifera.Genes as CoPLAC8-5,CoPLAC8-7,CoPLAC8-17,CoPLAC8-18,CoPLAC8-20 and CoPLAC8-23 may be related with Cu enrichment in C.oleifera kernel.
