摘要
目的:观察九味固金胶囊联合运动疗法对慢性阻塞性肺疾病(COPD)模型大鼠肺组织的病理改变及炎症因子IL-6、TNF-α水平的影响。方法:选取50只大鼠随机分为空白组、模型组、九味固金胶囊组、九味固金胶囊联合运动疗法组、氨茶碱组,每组各10只。采用烟熏联合气管内滴注脂多糖(LPS)方法建立COPD大鼠模型,造模成功后通过灌胃方式予各组大鼠相应药物的干预治疗。干预治疗周期结束后采用腹主动脉取血法处死全部大鼠,留取肺组织,用于相关检测,通过HE染色法进行病理检测,采用ELISA法检测大鼠肺组织中IL-6、TNF-α的浓度情况。结果:与模型组比较,九味固金胶囊组、氨茶碱组、九味固金胶囊联合运动疗法组大鼠的肺组织病变程度和炎症因子IL-6、TNF-α的浓度水平均有不同程度的降低,其中九味固金胶囊联合运动疗法组降低较明显(P﹤0.05)。结论:九味固金胶囊联合运动疗法能够下调慢阻肺模型大鼠的IL-6、TNF-α炎症因子浓度水平,并减轻肺组织的病理损伤,这可能是九味固金胶囊联合运动疗法能够改善慢阻肺患者临床症状及生活质量的作用机制之一。
Objective:To approach the effects of Jiuwei Gujin capsule and exercise therapy on pathological alterations and levels of inflammatory factors IL-6 and TNF-αin lung tissues of modeled rats with chronic obstructive pulmonary disease(COPD).Methods:50 rats were selected and randomly divided into blank group,model group,Jiuwei Gujin capsule group,Jiuwei Gujin capsule combined with exercise therapy group and aminophylline group,10 rats in each group.Rat model of COPD was established by methods of smoking and intratracheal instillation of lipopolysaccharide(LPS).The rats in each group were given corresponding drugs by gastric gavage after successful modeling,in order to intervene and treat.After the cycle of intervening treatment was ended,all rats were executed by blood collection form abdominal aorta;the lung tissues were retained for relevant tests;the pathological tests were carried out by HE staining;the concentrations of IL-6 and TNF-αin the lung tissues of rats were detected by ELISA.Results:Compared with the model group,the extent of pulmonary tissue lesions and the concentration levels of inflammatory factors IL-6 and TNF-αwere reduced to different degrees in Jiuwei Gujin capsule group,aminophylline group and Jiuwei Gujin capsule combined with exercise therapy group,and there was a more pronounced reduction in Jiuwei Gujin capsule combined with exercise therapy group(P﹤0.05).Conclusion:Jiuwei Gujin capsule combined with exercise therapy can down-regulate the concentrations of inflammatory factors IL-6 and TNF-αin modeled rats of COPD and decrease the pathological damage of lung tissue,which may be one of the mechanisms that Jiuwei Gujin capsule combined with exercise therapy can improve the clinical symptoms and the quality of life of patients with COPD.
