胶质-淋巴系统在Aβ清除中作用的研究进展

摘要 阿尔茨海默病(Alzheimer's disease,AD)是全球最常见的神经变性疾病,是痴呆的主要病因之一,目前仍未有十分有效的治疗方法。β-淀粉样蛋白(amyloidβ-protein,Aβ)生成与清除的不平衡被认为是AD的主要发病机制,早期增强脑内Aβ清除可以延迟AD的发生、发展。近年来,随着胶质-淋巴系统概念的提出与脑膜淋巴管的发现,中枢神经系统内液体的定向运输成为热点。胶质-淋巴系统在清除Aβ中发挥关键作用,有望成为对抗神经变性疾病的新方向。本综述旨在梳理当前关于大脑胶质-淋巴系统在AD治疗中作用的研究进展,探讨其在疾病管理中的潜在利用价值。 Alzheimer's disease(AD)is the most common neurodegenerative disease worldwide and a leading cause of dementia.Currently,there are no highly effective treatments for this condition.The imbalance between the production and clearance of Aβ(amyloidbeta)protein is considered a primary pathogenic mechanism of AD.Enhancing the clearance of Aβin the brain during the early stages can delay the onset and progression of AD.In recent years,with the introduction of the concept of the glymphatic system and the discovery of meningeal lymphatic vessels,the directional transport of fluids within the central nervous system has become a research hotspot.The glymphatic system plays a crucial role in clearing amyloid-beta protein and holds promise as a novel approach to combat neurodegenerative diseases.This review aims to summarize the current research progress on the role of the brain's glymphatic system in AD treatment and explore its potential applications in disease management.
