低氧与凋亡相关因子在牦牛肠道的差异性分析 被引量:1

摘要 为探究低氧参与细胞衰老凋亡的作用机制,以牦牛小肠组织(空肠、回肠和十二指肠)为研究对象,对低氧诱导因子1α(HIF-1α)及细胞凋亡相关因子B淋巴细胞瘤2(Bcl-2)、Bcl-2家族相关蛋白Bax(Bax)、半胱氨酸天冬氨酸特异性蛋白酶(Caspase-3)、血管内皮生长因子(VEGF)在各年龄段(新生、5月龄和成年)的小肠组织中不同的表达模式进行分析;对不同年龄段牦牛小肠组织进行苏木精-伊红染色(H&E)、过碘酸雪夫染色(PAS)、Masson三色染色和免疫组织化学染色分析。H&E以及特殊染色结果显示,各肠道间的组织学测量指标差异主要集中在十二指肠的肠绒毛和肠腺中。此外,5月龄牦牛空肠较十二指肠和回肠达到最大胶原纤维含量。免疫组化结果显示,低氧以及凋亡相关因子差异同样集中表达在十二指肠和空肠。低氧条件促使HIF-1α转录,激活凋亡因子诱导细胞凋亡,随年龄增加,牦牛逐步适应低氧环境,HIF-1α表达下降。促凋亡因子Bax和Caspase-3发挥功能时,抗凋亡因子Bcl-2和VEGF发挥保护作用,调控细胞衰老凋亡。低氧和各相关凋亡因子在5月龄的空肠中表达显著下降,除5月龄的牦牛空肠中VEGF表达显著升高。以上研究结果为后续探究牦牛肠道发育过程中HIF-1α、VEGF、Bax、Bcl-2和Caspase-3的表达分布规律提供基础数据,并为揭示凋亡相关因子在牦牛肠道中的低氧适应性提供参考资料。 To investigate the role of hypoxia in cell aging and apoptosis,small intestinal tissues(jejunum,ileum,and duodenum)of yaks were selected as materials.The differential expression of hypoxia-inducible factor 1α(HIF-1α),B-cell lymphoma 2(Bcl-2),Bcl-2 family-related protein Bax(Bax),caspase-3,and vascular endothelial growth factor(VEGF)in the tissues of different ages(newborn,5 months old,and adult)was analyzed.Immunohistochemical analyses were performed by collecting small intestine tissues from yaks of all ages with special stains including hematoxylin-eosin staining(H&E),povidone-iodine acid staining(PAS)and Masson trichrome staining(Masson).Results of H&E and special staining showed that the differences in histologic measures among the intestines were concentrated in the villi and glands of the duodenum,which was consistent with its role as the primary site of digestion.In addition,the highest collagen fiber content was observed in the jejunum of 5-months-old yaks.The results of immunohistochemistry showed that low-oxygen and apoptosis-related factors were differentially expressed in the duodenum as well.Hypoxia induced the transcription of HIF-1αactivates apoptosis factors and induced cell apoptosis.As the age increased,yaks gradually adapted to the hypoxic environment,leading to a decrease in the expression of HIF-1αdecreases.These findings provide basic data for further investigations into the distribution of HIF-1α,VEGF,Bax,Bcl-2 and Caspase-3 during the development of yak intestinal tract,and provide references for revealing the apoptosisrelated factors involved in the hypoxic adaptation of the yak intestinal tract.
