库仑滴定法快速测定葡萄糖氧化酶的活性

Rapid determination of glucose oxidase activity by coulometric titration method

摘要 基于库仑滴定法测定葡萄糖氧化酶的活性。采用葡萄糖氧化酶催化氧化β-D-葡萄糖定量生成过氧化氢后,通过电解生成的Ce^(4+)滴定过氧化氢,记录滴定时间,根据法拉第电解定律,测得葡萄糖氧化酶活性。结果表明:当葡萄糖氧化酶活性为0.33~2.67 U/mL时,其与滴定时间呈良好的线性关系,方法检测限为4.67×10^(-6) U/mL,加标回收率为95.8%~102.5%,相对标准偏差为0.9%~1.5%,结果准确度与精密度较好。在95%置信区间下,库仑滴定法与电位滴定法测定相同试样的结果无显著差异。库仑滴定法测定葡萄糖氧化酶活性的操作简便,检测时间可控,能在数分钟内完成检测,且干扰因素少,更有利于在相关产品检测中应用。 Glucose oxidase activity was determined by coulometric titration method.After using glucose oxidase to catalyze the oxidation of β-D-glucose to quantitatively generate hydrogen peroxide, the hydrogen peroxide was titrated with Ce^(4+)generated by electrolysis, and the titration time was recorded. According to Faraday's law of electrolysis, the activity of glucose oxidase was measured. The results showed that when the activity of glucose oxidase was 0.33-2.67 U/mL, it showed a good linear relationship with titration time. The detection limit of the method was 4.67×10^(-6) U/mL, the recovery rate was 95.8 %-102.5 %, and the relative standard deviation was 0.9 %-1.5 %. The accuracy and precision of the results were good. At the 95 % confidence interval, there was no significant difference in the results obtained by coulometric titration method and potentiometric titration method for the same sample. The coulometric titration method for determining glucose oxidase activity was easy to operate, with controllable detection time, and could be completed within a few minutes. It also had few interfering factors, which was beneficial for its application in the detection of related products.
