摘要
目前,丙烯酰胺是公认的主要引起神经毒性物质,在一些油炸和烧烤的淀粉类食品及高温加工的咖啡豆等中均可产生。向速溶咖啡样本中添加13C3标记的丙稀酰胺内标溶液,用水作提取溶剂,通过固相萃取柱净化后,采用液质联用法进行测定。根据基质的干扰和丙烯酰胺的极性,对流动相配比及柱效进行优选,最终确定选用C8型色谱柱和初始流动相水项占比大,增加石墨化碳净化降低基质干扰。方法定量限为10μg/kg,回收率为94.2%~103.5%,精密度为2.41%~2.79%,相关系数为0.999 6。该方法净化处理样品效果较好,且具有良好的准确度和灵敏度,重现性好,可达到对咖啡中丙烯酰胺含量测定的技术指标。
Acrylamide is currently recognized as the main neurotoxic substance,which can be produced in some fried and grilled starchy foods,as well as high-temperature processed coffee beans.Add the acrylamide internal standard solution labeled with 13C3 to the sample,use water as the extraction solvent,purify it through a solid-phase extraction column,and determination was performed by LC-MS.Based on the interference of the matrix and the polarity of acrylamide,the ratio of the mobile phase and the selection of the chromatographic column were optimized.Finally,it was determined to use a C8 chromatographic column with a larger ratio of water to the initial mobile phase,while increasing graphite carbon purification to reduce matrix interference.The quantitative limit of the method was 10μg/kg,recovery rate between 94.2%and 103.5%,precision range between 2.41%and 2.79%,correlation coefficient of 0.9996.This method had good accuracy and sensitivity,and could meet the detection requirements of acrylamide in coffee.
