摘要
为探讨猪流行性腹泻病毒(PEDV)在规模化养殖场环境中的分布规律,制定精准有效的猪场生物安全防控措施,采用实时荧光定量PCR方法,对采集自河南省信阳市某空置6个月猪场的366份环境样品进行了PEDV核酸检测。结果显示,该猪场环境样品中有124份PEDV核酸阳性,总体阳性率为33.88%;不同区域PEDV检出率由高到低依次为排污系统50.00%(4/8)、猪舍环境33.92%(116/342)、洗消中心25.00%(4/16);值得注意的是,洗消中心的减速带、猪舍环境内的漏粪地板及排污系统的粪沟检出率均为100%。结果表明,该猪场清洗消毒空置6个月后仍有大量PEDV核酸残留。总体来说,研究发现了猪场清洗消毒的部分盲区,讨论了病毒残留点和猪场建设规划,以及空气、饲料和环境等因素间的关系,指出了猪场现有清洗流程的漏洞,为猪场建立完善的生物安全防控体系提供了参考。
In order to investigate the distribution pattern of porcine epidemic diarrhea virus(PEDV)in the environment of large-scale swine farms,and to develop precise and effective biosecurity measures for swine farms,this study used the real-time fluorescent quantitative PCR method to detect 366 environmental samples collected from a vacant swine farm in Xinyang cty,Henan province,China.The nucleic acid test results revealed that some environmental samples in the swine farm were positive for PEDV,with the detection rates of PEDV from high to low in different areas as follows:50%(4/8)in the sewage system,33.9%(116/342)in the swine house environment,and 25%(4/16)in the cleaning and disinfection center.Of note,the detection rates for the speed bumps in the cleaning and disinfection center,the leaking manure floors in the swine house environment,and the manure ditch in the sewage system were all 100%.This indicated that a amount of PEDV nucleic acid residues remained in the swine farm even after it had been thoroughly cleaned and disinfected for 6 months.The study identified blind spots in the disinfection process of the swine farm,discussed the relationship between virus residues,swine farm and swine house construction planning,and various factors,and pointed out the loopholes in the existing cleaning procedures of the swine farm,providing valuable insights for establishing a comprehensive biosecurity and control system for swine farms.
