健康口腔分离细菌对牙膏中抗菌防腐剂的敏感性分析

Susceptibility of Bacteria Isolated from Healthy Oral Cavity to Antibacterial Preservatives in Toothpaste

摘要 采用基质辅助激光解吸电离飞行时间质谱(MALDI-TOF MS)和16S rRNA序列鉴定对有氧条件下分离的29株口腔菌进行种属分析。通过测定29株口腔菌的最低抑菌浓度(MIC),分析牙膏产品中6种常用防腐剂的体外抗菌效果。种属鉴定结果表明,29个口腔菌株为链球菌属(55.17%,16/29)、奈瑟氏菌属(31.03%,9/29)和葡萄球菌属(13.79%,4/29)。结果显示,GB 22115—2008规定的4-羟基苯甲酸丙酯的最大允许使用浓度明显高于29株口腔分离株的MIC(约8~64倍)。结果表明牙膏产品中的某些防腐剂可能会抑制或杀死口腔菌,从而对口腔菌群造成不良影响。防腐剂最大允许剂量的确定不仅应基于毒理学数据,还应考虑口腔微生物的抗菌敏感性。 Matrix-assisted laser desorption ionization time-of-flight mass spectrometry(MALDI-TOF MS)and 16S rRNA sequence identification were used to analyze the species of 29 oral bacteria isolated under aerobic conditions.The in vitro antibacterial effects of 6 commonly used preservatives in toothpaste products were analyzed by determining the minimum inhibitory concentration(MIC)of 29 oral bacteria.The species identification results showed that 29 oral bacterial strains were Streptococcus(55.17%,16/29),Neisseria(31.03%,9/29)and Staphylococcus(13.79%,4/29).The results showed that the maximum allowable use concentration of propyl 4-hydroxybenzoate in GB 22115—2008 was significantly higher than the MIC of 29 oral isolates(about 8 to 64 times).The results indicated that some preservatives in toothpaste products might inhibit or kill oral bacteria,thus having adverse effects on oral flora.The determination of the maximum allowable dose of preservatives should not only be based on toxicological data,but also consider the antibacterial sensitivity of oral microorganisms.
