‘Kitaake’水稻MAPK级联基因家族鉴定及辐射后表达分析

Identification of MAPK cascade gene family and its expression analysis after radiation in rice variety‘Kitaake’

摘要 本研究以水稻粳稻品种‘Kitaake’(Oryza sativa ssp.japonica cv.Kitaake)为研究对象,鉴定并分析了丝裂原活化蛋白激酶(mitogen-activated protein kinase,MAPK)级联基因家族成员,并通过转录组测序分析了‘Kitaake’幼苗经高能重离子束和X射线辐射处理后MAPK级联基因家族成员的表达模式。在‘Kitaake’中共鉴定到17个OskMAPK、12个OskMAPKK和17个OskMAPKKK基因,其编码蛋白的分子质量分别在24243.00~69400.23、32736.94~60054.98和36967.51~150503.80 Da之间。在基因上游存在多个与生长发育、植物激素、胁迫响应相关的启动子调控元件。‘Kitaake’和‘日本晴’(O.sativa ssp.japonica cv.Nipponbare)的MAPK级联家族间共有36对共线性关系。重离子束和X射线辐射‘Kitaake’幼苗后分别有15、17个OskMAPK级联基因表达量显著变化。以上结果揭示了‘Kitaake’水稻的MAPK级联基因和蛋白特征,以及水稻幼苗在两种辐射处理后不同部位MAPK级联信号调控的特点,为MAPK级联基因在辐射响应中的功能研究提供基础依据。 This study identified and analyzed MAPK(mitogen-activated protein kinase)cascade gene family members in rice variety‘Kitaake’(Oryza sativa ssp.japonica cv.Kitaake).We analyzed the expression patterns of MAPK cascade genes of‘Kitaake’after high-energy heavy ion beam(HIB)and X-ray irradiations through transcriptome sequencing.The results showed that there are 17 OskMAPKs,12 OskMAPKKs,and 17 OskMAPKKKs in‘Kitaake’.The molecular masses of the proteins they encode were 4243.00 to 69400.23,32736.94 to 60054.98,and 36967.51 to 150503.80 Da,respectively.MAPK cascade genes generally contains multiple promoter regulatory elements related to growth and development,plant hormones,and stress responses.There are 36 pairs of collinear relationships between‘Kitaake’and‘Nipponbare’(O.sativa ssp.japonica cv.Nipponbare).After HIB and X-ray irradiation,there were 15 and 17 Osk-MAPK cascade genes in‘Kitaake’seedlings showed significant changes in expression levels,respectively.These results reveal the characteristics of MAPK cascade genes and their proteins of‘Kitaake’,as well as the expression regulation characteristics of these genes in different parts of seedlings after irradiation.This study could provide a foundation for the functional research of MAPK cascade genes’response to radiations.
