针刺对抑郁模型大鼠海马nNOS与PSD-95共表达的影响 被引量:1

Effect of Acupuncture on Coexpressions of nNOS and PSD-95 in Hippocampus of Depression Model Rats

摘要 目的观察针刺治疗抑郁模型大鼠对海马中神经元型一氧化氮合酶(neuronal nitric oxide synthase,nNOS)与突触后致密蛋白-95(postsynaptic density-95,PSD-95)共表达的影响。方法选择SPF级大鼠50只,随机分为正常组、模型组(腹腔注射利血平0.2 mg·kg^(-1)·d^(-1))、针刺组、ZL006组、针刺联合ANA-12组、8-OH-DPAT组共6组,正常组和模型组9只,余组各8只,观察体质量和肛温变化;采用旷场实验、糖水偏嗜实验、强迫游泳实验观察大鼠抑郁样行为;采用HE染色观察海马神经元形态变化;采用免疫荧光双标染色观察海马CA1区、CA3区神经元中nNOS与PSD-95、酪氨酸蛋白激酶受体B(tyrosine kinase receptor B,TrkB)与PSD-95的表达情况。结果海马CA1区、CA3区神经元中nNOS与PSD-95、TrkB与PSD-95存在共表达,与模型组比较,针刺组和ZL006组抑郁样行为均显著改善(P<0.05,P<0.01,P<0.001),CA3区nNOS与PSD-95的共表达水平显著下调(P<0.05,P<0.01),CA3区TrkB与PSD-95的共表达水平显著上调(P<0.05),海马神经元减少和损伤显著改善,针刺组海马CA1区nNOS的表达水平显著下调(P<0.01),CA1区TrkB的表达水平显著上调(P<0.01),8-OH-DPAT组CA1区nNOS的表达水平显著下调(P<0.01);与针刺组比较,ANA-12组CA3区nNOS与PSD-95的共表达水平显著上调(P<0.05),以上差异均有统计学意义。结论针刺调节少阳枢机,可能通过上调海马CA3区神经元中nNOS与PSD-95的共表达水平参与发挥抗抑郁作用,该作用机制可能与CA3区TrkB受体有关。 Objective To observe the effect of acupuncture treatment on coexpression of neuronal nitric oxide synthase(nNOS)and postsynaptic dense-95(PSD-95)in the hippocampus of depression model rats.Method Fifty SPF grade rats were randomly divided into six groups:normal group,model group(intraperitoneal injection of 0.2 mg·kg^(-1)·d^(-1)of reserpine),acupuncture group,ZL006 group,acupuncture combined with ANA-12 group,8-OH-DPAT group.There were 9 rats in the normal group and model group,and 8 rats in each of the remaining groups.The changes in body weight and anal temperature were observed.Open field test,sucrose preference test and forced swimming test were applied to observe depression-like behaviors in rats.The morphological changes of hippocampal neurons were observe by HE staining.Immunofluorescence double staining was used to observe the positive expression of nNOS and PSD-95,as well as tyrosine kinase receptor B(TrkB)and PSD-95 in neurons of the hippocampal CA1 and CA3 regions.Result There were coexpressions of nNOS and PSD-95 as well as TrkB and PSD-95 in neurons of the hippocampal CA1 and CA3 regions.Compared with those in the model group,depression-like behaviors were significantly improved in the acupuncture group and ZL006 group(P<0.05,P<0.01,P<0.001).The coexpression levels of nNOS and PSD-95 in the CA3 region were significantly down-regulated(P<0.05,P<0.01),and the coexpression levels of TrkB and PSD-95 in the CA3 region were significantly up-regulated(P<0.05).The reduction and damage of hippocampal neurons were significantly improved.The expression levels of nNOS in the hippocampal CA1 region were significantly down regulated(P<0.01)and the expression levels of TrkB in the CA1 region were significantly up-regulated(P<0.01)in the acupuncture group.The expression level of nNOS in the CA1 region of the 8-OH-DPAT group was significantly down-regulated(P<0.01).Compared with those in the acupuncture group,the coexpression levels of nNOS and PSD-95 in the CA3 region of the ANA-12 group were significantly up-regulated(P<0.05),and the above differences were statistically significant.Conclusion Acupuncture regulates Shaoyang pivot,which may participate in the antidepressant effect by up-regulating the coexpression levels of nNOS and PSD-95 in hippocampal CA3 neurons,and this mechanism of action may be related to TrkB receptors in the CA3 region.
