长牡蛎牛磺酸转运蛋白Cg TauT基因的结构和表达特征

摘要 为探究长牡蛎(Crassostrea gigas)(湿质量为155.0 g±5.0 g)牛磺酸转运蛋白(taurine transporter,TauT)基因家族成员组成及其表达特征,采用全基因组筛查策略对长牡蛎牛磺酸转运蛋白基因进行鉴定,并利用qRT-PCR对其组织分布模式和牛磺酸处理下的表达特征进行分析。结果表明:长牡蛎基因组中存在4个Cg TauT基因,均包含1个SNF结构域和12个跨膜结构域;4个Cg TauT基因在长牡蛎各组织中呈组成性表达,其中Cg TauT1在闭壳肌中表达量最高,Cg TauT2、Cg TauT3和Cg TauT4在外套膜中表达量最高,不同组织中Cg TauT基因表达量与牛磺酸含量无显著相关性;牛磺酸浸泡处理下4个Cg TauT基因在外套膜中呈现不同的时序表达模式,其中Cg TauT1的表达量在处理3 h后率先升高,Cg TauT3的表达量随处理时间延长呈逐渐升高趋势,Cg TauT4的表达量随处理时间延长呈先升高后降低的趋势。本研究结果为深入探究Cg TauT的生理功能和长牡蛎的分子机制提供了科学参考。 To investigate the composition and expression characteristics of the taurine transporter(Cg TauT)gene family in Crassostrea gigas,a genome-wide screening strategy was used to identify taurine transporter genes,and the qRT-PCR was used to analyse their tissue distribution pattern and to expression characteristics after taurine treatment.The results revealed that C.gigas contains four Cg TauT genes,with each containing an SNF structural domain and 12 transmembrane domains.The four Cg TauT genes were constitutively expressed in the various tissues of C.gigas,with Cg TauT1 showing the highest expression in the adductor muscle,and Cg TauT2,Cg TauT3 and Cg TauT4 showing the highest expression in the mantle.There were no significant correlations between the expression levels of the four Cg TauT genes and the taurine contents of the tissues.Under taurine immersion treatment,the four Cg TauT genes exhibited different temporal expression profiles in the mantle of C.gigas.The expression level of Cg TauT1 increased after three hours of treatment.With prolonged taurine treatment,the mRNA expression level of Cg TauT3 increased gradually,while that of Cg TauT4 first increased and then decreased.These findings provide a theoretical foundation for further exploration of the physiological functions of Cg TauTs and the molecular mechanisms underlying taurine accumulation in C.gigas.
