不同提取工艺的丹参制剂对气滞血瘀模型大鼠的药理作用影响分析 被引量:1

Effects of different extraction processes of Salvia miltiorrhiza preparations on the pharmacological effects in rats with Qi stagnation and blood stasis model

摘要 目的探讨不同提取工艺制备下的丹参膏、丹参片对气滞血瘀模型大鼠的主要药效的差别。方法将48只SD雌性大鼠随机分为8组,每组6只,分别为空白组、模型组、复方丹参片组(1.10 g/kg)、丹参片A组(0.81 g/kg)、丹参片B组(0.81 g/kg,以生药量计)、丹参膏高剂量组(3.24 g/kg)、丹参膏中剂量组(1.62 g/kg)、丹参膏低剂量组(0.81 g/kg);给药组每天进行预防性给药,给药后除空白组外其余各组大鼠均采取夹尾和冰水浴刺激复合注射盐酸肾上腺素制备气滞血瘀模型,每天观察大鼠症状;比较各组大鼠血瘀证证候积分、升主动脉与心肌组织的病理学变化、凝血四项(APTT、PT、TT、FIB)、一氧化氮(NO)、内皮素1(ET-1)、前列腺素(PGE2、PGF2α)、缩宫素(OT)、雌二醇(E_(2))含量。结果与空白组比较,模型组PT、TT均减短(P<0.05),证候积分以及FIB、NO、ET-1、PGE2、PGF2α、OT、E_(2)含量均明显异常(P<0.05),升主动脉和心脏均受到严重损坏。与模型组比较,给药组证候积分均下降,升主动脉与心脏损伤程度均得到改善,除丹参片A组外差异均有统计学意义(P均<0.05);各给药组NO、ET-1、PGF2α、OT、E_(2)水平显著得到改善(P均<0.05);各给药组均能够延长PT,复方丹参片、丹参膏低剂量组、丹参片B组有统计学意义(P<0.05);复方丹参片组,丹参膏低剂量组,丹参片A、B组均能延长TT,但仅丹参片B组差异有统计学意义(P<0.05);仅有丹参片B组能显著延长APTT(P<0.05);各给药组均未能改善FIB水平(P>0.05);丹参片A、B组能显著提高PGE2水平(P<0.05)。结论丹参膏与丹参片均能调节气滞血瘀型大鼠的凝血系统、心血管系统、妇科系统,丹参膏的治疗效果与剂量密切相关,随剂量增大疗效降低;中医认为膏剂以滋养膏润为长,片剂以速效方便为长,临床应用可根据疾病的轻重缓急做出合理的选择,且应注意选择合适的厂家,提高药物的效度。 Objective To evaluate the differences in the main medicinal effects of Danshen Gao and Danshen Pian on rats in the model of Qi stagnation and blood stasis under the preparation of different extraction processes.Methods 48 SD female rats were randomly divided into 8 groups,with 6 rats in each group which the control group,the model group,the compound Danshen Pian group(1.10 g/kg),Danshen Pian group A(0.81 g/kg),Danshen Pian group B(0.81 g/kg),the high-dose Danshen Gao group(1.62 g/kg),and the low-dose Danshen Gao group(0.81 g/kg),The drug administration group received preventive medication daily.After dosing,all groups except the control group underwent tail clamping and ice water bath stimulation combined intravenous injection of adrenaline hydrochloride to prepare a Qi stagnation and blood stasis model.Rats were observed for symptoms and compared with each group for blood stasis evidence points,pathological changes in ascending aorta and myocardial tissues,four items of coagulation(APTT,PT,TT,FIB),nitric oxide(NO),endothelin 1(ET-1),prostaglandins(PGE2,PGF2α),oxytocin(OT),and estradiol(E_(2))level.Results Danshen Gao,Danshen Pian and Fufang Dashen Pian given were all able to reduce the evidence points and improve the damaged ascending aorta and cardiac injury,except for Pian(tablet)group,which there were statistically significant differences(P<0.05).Danshen Gao,Danshen Pian,and Fufang Danshen Pian were all able to significantly improve the levels of NO,ET-1,PGF2α,OT,and E_(2)(P<0.05).Danshen Gao,Danshen Pian,and Fufang Dashen Pian were all able to prolong PT,and there were statistically significant differences between Fufang Danshen Pian,Danshen Gao low dose,and Danshen Pian 2(P<0.05).The group of Fufang Dashen Pian,the group of Danshen Gao low dose,and the groups of Danshen Pian A and B were able to prolong TT,and there were statistically significant differences in Danshen Pian B(P<0.05).The group of Danshen Pian B was able to prolong APTT significantly(P<0.05).The groups of all drug administration failed to improve the levels of FIB.The groups of Danshen Pian A and B were able to significantly increase the level of PGE2(P<0.05).Conclusion Both Danshen Gao and Danshen Pian can regulate the coagulation system,cardiovascular system and gynaecological system of rats with Qi stagnation and blood stasis.The therapeutic effect of Danshen Gao is closely related to the dosage,and the therapeutic effect decreases as the dosage increases.Chinese medicine believes that creams to play the role of nourishing the body mainly,tablets are better for rapid and convenient action.Clinical application can be based on the severity of the disease to make a reasonable choice,and should pay attention to the selection of the right manufacturer,to improve the efficacy of the drug.
