基于UPLC-Q-Exactive Orbitrap-MS分析皱皮木瓜与光皮木瓜化学成分

Analysis of chemical constituents of Chaenomeles speciosa and Chaenomeles sinensis based on UPLC-Q-Exactive Orbitrap-MS

摘要 目的利用超高效液相色谱-四极杆-静电场轨道阱高分辨质谱法(UPLC-Q-Exactive Orbitrap-MS)分析皱皮木瓜、光皮木瓜化学成分,为光皮木瓜植物资源开发利用提供参考。方法采用Halo C 18色谱柱(2.1 mm×100 mm,2.7μm),以0.05%甲酸(A)-甲酸乙腈(B)为流动相梯度洗脱,体积流量0.3 mL·min^(-1),柱温40℃,进样量3μL,质谱选用ESI源,分别以正负离子模式切换扫描采集数据,根据精准分子量、二级质谱碎片离子信息,结合文献报道、自建数据库及裂解规律等对化学成分进行鉴定。结果光皮木瓜共鉴定成分42种,其中有机酸及其衍生物12种、黄酮及香豆素类成分23种和萜及其他类成分7种;皱皮木瓜鉴定成分36种,其中有机酸及其衍生物14种、黄酮及香豆素类成分14种、萜及其他类成分8种;二者相同成分有25种。结论光皮木瓜较皱皮木瓜含有种类更多的化学成分,二者相同成分占皱皮木瓜成分种类的69.44%,提示光皮木瓜具有重要的药用开发潜力。 Objective The aim of this study was to analyze the chemical compositions of Chaenomeles speciosa and Chaenomeles sinensisusing UPLC-Q-Exactive Orbitrap-MS and compare the composition differences of two species.Methods The separation was performed on a Halo C 18 column(2.1 mm×100 mm,2.7μm)with a gradient elution of 0.05%formic acid(A)-formic acid acetonitrile(B)as the mobile phase.Flow rate was 0.3 mL·min^(-1) and column temperature was 40℃ with an injection volume 3μL.The mass spectrometry was performed using an ESI source,and the data were collected by switching the positive and negative ions modes,respectively.The data were collected by switching the scanning mode of positive and negative ions,and the components were identified according to the precise molecular weight,secondary mass spectrometry fragmentation ion information,combined with the literature reports,self-constructed database,and cleavage pattern.Results A total of 42 constituents were identified in Chaenomeles sinensis,including 12 organic acids and their derivatives,23 flavonoids and coumarins,and seven terpenoids and others.Meanwhile,36 constituents were identified in Chaenomeles speciosa,including 14 organic acids and their derivatives,14 flavonoids and coumarins,and eight terpenoids and others;Among these,both have 25 indentical constituents.Conclusion There were more chemical components in Chaenomeles sinensis than in Chaenomeles speciosa,and the same components accounted for 69.44%of the components in Chaenomeles speciosa,suggesting that Chaenomeles sinensis has significant medicinal value,with more tremendous potential for development and utilization.
