火麻仁对SW620结肠癌细胞ATAD2表达的抑制作用及化学成分研究

摘要 目的:探究火麻仁对SW620结肠癌细胞的抑制作用及其化学成分。方法:以RT-qPCR技术为检测手段,考察火麻仁乙醇提取液对结肠癌SW620细胞三磷酸腺苷家族蛋白2(ATPase family AAA domain-containing protein 2,ATAD2)基因表达水平的影响;采用超高效液相色谱/四级杆-静电场轨道阱质谱,分别对液相色谱流动相种类、柱温、梯度洗脱条件,质谱分辨率、喷雾电压等条件进行优化,对火麻仁乙醇提取液中的化学成分进行定性检测。结果:火麻仁乙醇提取液可抑制结肠癌SW620细胞ATAD2的基因表达;在火麻仁提取液中共定性检测到22个化学成分。结论:火麻仁乙醇提取液具有潜在的抗结肠癌活性,其潜在活性成分包括脂肪酸类、黄酮类及生物碱类。 Objective:To explore the inhibitory effect of Cannabis Fructus(CF)on colon cancer SW620 cells as well as its components.Methods:The effect of CF on the ATAD2 gene expression in colon cancer SW620 cells was investigated by real-time fluorescent quantitative reverse transcription polymerase chain reaction(RT-qPCR).Ultra-high performance liquid chromatography/quadrupole-exactive orbitrap mass spectrometry(UHPLC/Q-Exactive Orbitrap MS)was used to detect the components in CF after optimizing the detection conditions including liquid chromatography mobile phase,column temperature,gradient elution conditions,mass spectrometry resolution,spray voltage,etc.Results:The extraction of CF could inhibit the gene expression of ATAD2 in colon cancer SW620 cells.There were total 22 components being qualified in CF.Conclusion:The CF extract possessed potential anti-colon cancer activity,and its active components include fatty acids,flavonoids and alkaloids.
