摘要
基质金属蛋白酶‑3(matrix metallopeptidase 3,MMP‑3)在中枢神经系统中具有广泛的生物学功能,尤其在损伤修复及神经病理性疾病的发生、发展中发挥关键作用。MMP‑3通过降解细胞外基质(extracellular matrix,ECM)成分,促进轴突和髓鞘再生,并抑制神经再生抑制因子。此外,MMP‑3还通过调节血脑屏障(blood‑brain barrier,BBB)的通透性和调控神经炎症,在帕金森病(parkinson's disease,PD)和阿尔茨海默病(alzheimer's disease,AD)等神经退行性疾病的病理进程中发挥重要作用。值得注意的是,MMP‑3的异常表达与脑肿瘤的侵袭性及恶性程度密切相关。本文综述了MMP‑3在这些中枢神经系统疾病中的作用机制,并探讨了其作为潜在治疗靶点的应用前景。未来的研究应着重于深入解析MMP‑3的分子调控机制,以推动靶向治疗策略的开发,提高相关疾病的治疗效果。
Matrix metallopeptidase 3(MMP‑3)has a wide range of biological functions in the central nervous system(CNS),particularly playing a key role in injury repair and the development of neuropathological diseases.MMP‑3 promotes axonal and myelin regeneration by degrading extracellular matrix(ECM)components and inhibiting nerve regeneration inhibitors.Additionally,MMP‑3 regulates blood‑brain barrier(BBB)permeability and modulates neuroinflammation,contributing to the pathological processes of neurodegenerative diseases such as Parkinson's disease(PD)and Alzheimer's disease(AD).Notably,abnormal MMP‑3 expression is closely related to the invasiveness and malignancy of brain tumors.This review summarizes the mechanisms by which MMP‑3 operates in these CNS diseases and discusses its potential as a therapeutic target.Future research should focus on in‑depth analysis of the molecular regulatory mechanisms of MMP‑3 to advance the development of targeted therapeutic strategies and improve treatment outcomes for related diseases.
