灯盏花素对冠心病血瘀证大鼠血浆N^(1)-甲基烟酰胺水平的影响

Effects of Breviscapine on Plasma Level of N^(1)-methylnicotinamide in Rats with Blood Stasis Syndrome of Coronary Artery Disease

摘要 目的:研究灯盏花素对冠心病血瘀证大鼠的治疗作用及血浆N^(1)-甲基烟酰胺(MNA)水平的影响。方法:通过腹腔注射维生素D3结合高脂饮食制备大鼠血瘀证模型,在此基础上采用皮下多点连续注射异丙肾上腺素制备冠心病血瘀证模型。SD大鼠随机分为对照组、模型组、灯盏花素低剂量组(8 mg·kg^(-1))和灯盏花素高剂量组(24 mg·kg^(-1)),每组8只。尾静脉注射给药,每天1次,连续7 d,对照组和模型组注射等体积生理盐水。检测各组血脂、乳酸脱氢酶(LDH)含量及血浆MNA浓度,进行血液流变学、心电图及心脏组织病理学检查。结果:与对照组比较,模型组血脂、LDH、MNA水平明显改变(P<0.05或P<0.01),心电图T波出现倒置、低平或抬高,心肌细胞排列紊乱,伴有炎性细胞浸润;与模型组比较,灯盏花素高剂量组血脂、LDH、MNA水平明显改变(P<0.05),T波变化和心肌病理损伤得到明显改善。结论:灯盏花素能够改善冠心病血瘀症大鼠心肌缺血,对冠心病血瘀症的防治作用可能与降低血浆MNA水平有关。 Objective:To investigate the effects of breviscapine(BRE)on blood stasis syndrome of coronary artery disease(BSS-CAD)and plasma level of N^(1)-methylnicotinamide(MNA)in Sprague-Dawley(SD)rats.Methods:The rat model of blood stasis syndrome was prepared by intraperitoneal injections of vitamin D3 and high-fat feeding,then BSS-CAD model was prepared by continuous subcutaneous injec-tions of isoproterenol.The rats were randomly divided into control group,model group,low dose and high dose breviscapine groups(8 mg·kg^(-1),24 mg·kg^(-1)),8 rats in each group.Breviscapine were injected via tail vein once a day for 7 days.Rats in the control group and model groups were given equal volume normal saline.Results:Compared with the control group,blood lipid and MNA levels were significantly changed(P<0.01 or P<0.05);T wave was inverted,flat or elevated;myocardial cells were disordered and accom-panied by inflammatory cell infiltration in the model groups.Compared with the model group,blood lipid and MNA levels were significantly reduced(P<0.05);the changes of T wave and myocardial pathological injury were significantly improved in the high dose breviscapine group.Conclusion:Breviscapine signifi-cantly improves myocardial ischemia in BSS-CAD rats,and the preventive and therapeutic effects on BSS-CAD may be related to reducing the plasma level of MNA.
