生物堆肥对土壤中芥子气模拟剂2-CEES降解效果的影响研究

摘要 本文旨在探究生物堆肥对土壤中芥子气模拟剂2-氯乙基乙基硫醚(2-Chloroethyl Ethyl Sulfide,2-CEES)降解效果的影响,为芥子气受染土壤的生态修复提供科学依据和实践指导。采用气相色谱技术建立了一种快速分析土壤中2-CEES残留量的方法,并利用六通道堆肥呼吸仪和16S rRNA测序考察了堆肥温度和生物强化措施对2-CEES降解行为的影响。结果显示,LB肉汤刺激能有效促进2-CEES在土壤中的降解,且中温堆肥结合生物强化的方法降解效果最佳。2-CEES污染显著抑制了土壤呼吸,改变了土壤微生物群落结构的均匀度和丰富度,酸杆菌门和绿弯菌门的相对丰度分别增加了7.23%~103.20%和17.69%~116.99%,放线菌门的相对丰度下降了13.54%~53.29%,且与2-CEES降解率高度关联(P<0.01)。结果表明,生物堆肥技术能提高土壤中2-CEES的降解率,其作用机理与微生物群落结构的改变有关,相关成果可为芥子气污染土壤的生物修复提供新的思路和方法。 The purpose of this study was to explore the effect of biocompost on the degradation effect of mustard gas simulant 2-chloroethyl ethyl sulfide(2-CEES)in soil to provide a scientific basis and practical guidance for the ecological restoration of mustard gas-contaminated soil.A rapid method for the analysis of 2-CEES residues in soil was established by gas chromatography.The effects of biocomposting temperature and bioaugmentation measures on the degradation behaviour of 2-CEES were investigated by soil composter and 16S rRNA.The results showed that LB broth stimulation could effectively promote the degradation of 2-CEES in soil,and the degradation effect of medium-temperature composting combined with bioaugmentation was the best.2-CEES pollution significantly inhibited soil respiration and changed the uniformity and richness of soil microbial community structure,and the relative abundance of Acidobacteria and Chloroflexi increased by 7.23%-103.20%and 17.69%-116.99%,respectively.The relative abundance of Actinobacteriota decreased by 13.54%-53.29%and was highly correlated with the degradation rate of 2-CEES(P<0.01).The results showed that composting technology could improve the degradation efficiency of 2-CEES in soil,and its mechanism was related to the change of microbial community structure.The relevant results were expected to provide new ideas and methods for the bioremediation of mustard gas-contaminated soil.
