柠檬酸改性的羟基磷灰石纳米片抗病毒效果及作用方式

摘要 为探究柠檬酸改性的羟基磷灰石纳米片(HA-Cit)对猪伪狂犬病毒(PRV)和猪传染性胃肠炎病毒(TGEV)的抑制活性及其作用方式,以CaCl_(2)、Na_(2)HPO_(4)、柠檬酸为原料,采用水热法合成HA-Cit,通过高分辨透射电镜、激光粒度仪、红外光谱仪、X射线衍射仪对HA-Cit进行图谱分析,并应用CCK-8法检测HA-Cit细胞毒性、病毒空斑法检测HA-Cit抗病毒活性及对病毒的作用方式。结果表明:相较于纯相的羟基磷灰石纳米棒(HA),HA-Cit为片状,粒径增加,拥有更强的负电性和分散性,含有柠檬酸特征官能团,结晶度降低;HACit具有更高的生物相容性,对两种病毒的治疗指数(I_(S))均显著高于HA,其抗病毒作用方式为直接灭活病毒及抑制病毒的复制和释放。 In order to explore the inhibitory activity and action mode of citric acid-modified hydroxyapatite nanosheets(HA-Cit)on porcine pseudorabies virus(PRV)and porcine infectious gastroenteritis virus(TGEV),CaCl_(2),Na_(2)HPO_(4) and citric acid were used as raw materials to prepare HA-Cit by hydrothermal method,and HA-Cit was characterized by high-resolution transmission electron microscopy,laser particle size analyzer,infrared spectrometer and X-ray diffractometer.Subsequently,the cytotoxicity of HA-Cit was detected by CCK-8 method,and the antiviral activity and action mode on viruses were detected by viral plaque method.The results showed that compared with the pure phase hydroxyapatite nanorods(HA),HA-Cit was flaky,with increased particle size,higher electronegative and dispersible properties,and contained characteristic functional groups of citric acid,with reduced crystallinity.Furthermore,HA-Cit had better biocompatibility and the selection index(I_(S))of both viruses was significantly higher than that of HA,and its antiviral mode was to directly inactivate the virus and inhibit the replication and release of the virus.
