摘要
CdTe/CdS/SiO_(2)量子点因其明亮的发光特性和优异的光稳定性而在生物领域具有很大的应用潜力,但其生物安全问题仍不清楚。该研究通过低剂量(0.4 mg/kg)和高剂量(4 mg/kg)的单次腹腔注射来评估CdTe/CdS/SiO_(2)量子点对小鼠的亚急性毒性,同时监测42 d的体重、脏器系数、血细胞计数、氧化指数、组织病理学变化和生物分布。结果表明,注射CdTe/CdS/SiO_(2)量子点后,小鼠体重和脏器系数无显著差异。荧光观察显示,CdTe/CdS/SiO_(2)量子点在器官中的早期积累为肺>脾>肝>肾>心,在分布上表现出明显的偏好。电感耦合等离子体质谱分析显示,CdTe/CdS/SiO_(2)量子点注射后在小鼠主要器官积累42 d,且早中期主要分布在脾和肺,后期主要分布在肝和肾。量子点处理组与对照组间的大部分血细胞计数和氧化指数均无显著性差异。此外,与对照组相比,经CdTe/CdS/SiO_(2)量子点处理的小鼠未发现明显的组织病理学异常。简而言之,CdTe/CdS/SiO_(2)量子点在小鼠体内体现出了良好的生物相容性,这些发现为量子点的安全应用提供了有价值的数据。
CdTe/CdS/SiO_(2) quantum dots have great potential for biological applications due to their bright luminescent properties and excellent photostability,but their biosafety issues remain unclear.In this study,the subacute toxicity of CdTe/CdS/SiO_(2) quantum dots in mice was evaluated by single intraperitoneal injections at low(0.4 mg/kg)and high(4 mg/kg)doses.The body weights,organ coefficient,blood cell counts,oxidative indices,histopathological changes,and biodistribution were continually monitored for 42 days.Results showed that there was no significant difference in body weight and organ coefficient of mice after injection of CdTe/CdS/SiO_(2) quantum dots.Fluorescence observation showed that the early accumulation of CdTe/CdS/SiO_(2) quantum dots in organs was lung>spleen>liver>kidney>heart,which showed obvious preference in distribution.Inductively coupled plasma mass spectrometry analysis showed that CdTe/CdS/SiO_(2) quantum dots were accumulated in the major organs of the mice for 42 days after the injection,and they were mainly distributed in the spleen and lungs in the early and middle stages,and in the liver and kidney in the later stages.Most of the blood cell counts and oxidative indices were not significantly different between the quantum dot-treated group and the control group.In addition,no significant histopathological abnormalities were found in the CdTe/CdS/SiO_(2) quantum dot-treated mice compared to the control group.In short,CdTe/CdS/SiO_(2) quantum dots demonstrated good biocompatibility in mice.These findings provide valuable data for the safe application of quantum dots.
