摘要 目的:提升蜂王浆的质量标准。方法:采用薄层色谱(TLC)法对蜂王浆中脯氨酸、赖氨酸、谷氨酸进行定性鉴别;采用80℃烘干法测定蜂王浆水分,采用高效液相色谱法测定10-羟基2-癸烯酸的含量,色谱柱为Agilent ZORBAX SB-C_(18)(250 mm×4.6 mm,5μm),流动相:乙腈-水(22∶78);流速1 mL/min;检测波长212 nm;柱温:30℃;对照品进样量5μL,样品进样量10μL。结果:脯氨酸、赖氨酸、谷氨酸的TLC图斑点清晰,阴性对照无干扰。10-羟基-2-癸烯酸的进样量分别在0.0104~1.0402 mg/mL范围内与峰面积线性关系良好;精密度、稳定性、重复性试验的RSD均小于2.0%;平均加样回收率为102.0%,RSD为1.8%;13批样品平均含量为1.906%。结论:所建立的方法专属性较强,重复性好,可用于蜂王浆的质量控制。 Objective:To improve the quality standard of royal jelly.Methods:The qualitative identification of proline acid,lysine and histidine in royal jelly was carried out by thin-layer chromatography(TLC);the moisture content of royal jelly was determined by drying at 80℃,and the content of 10-hydroxydec-2-enoic acid was determined by high-performance liquid chromatography(HPLC)on an Agilent ZORBAX SB-C_(18) column(250 mm×4.6 mm,5μm)with acetonitrile-water(22∶78)as a mobile phase;flow rate of 1 mL/min;detection wavelength of 212 nm;column temperature:30℃;the control product was injected into the column at a flow rate of 1 mL/min.The mobile phase was acetonitrile-water(22∶78)at a flow rate of 1 mL/min;the detection wavelength was 212 nm;the column temperature was 30℃;the injection volume of the control was 5μL and that of the sample was 10μL.The TLC plots of the four amino acids,glutamic acid,proline,lysine and histidine,were clear with no interference from the negative control.Results:The TLC plots of proline acid,lysine and histidine were clear,and there was no interference with the negative control.The linear relationship between the sample volume of 10-hydroxy-2-decenoic acid and the peak area was good in the range of 0.0104 mg/mL-1.0402 mg/mL,respectively.The RSD of the precision,stability,and reproducibility tests were all less than 2.0%,and the average recoveries of the mascot were 102.0%with the RSD of 1.8%,and the average content of the 13 batches of samples was 1.906%.Conclusion:The established method is highly specific and reproducible,and can be used to control the quality of royal jelly.
