摘要
为了解山东地区鸭圆环病毒(DuCV)的流行情况并探究其分子特征,试验从山东省部分地区疑似感染鸭圆环病毒的病鸭中采集样品进行病原检测,初步判定为鸭圆环病毒感染,进一步采用常规PCR方法扩增出4株DuCV的全基因组序列,并进行遗传进化和分子特征分析。结果显示:4株病毒的全基因组序列相似性在96.6%~99.6%之间;4株病毒与代表株DQ100076位于同一分支,相似性在97%以上,属于DuCV-1b亚型;4株病毒的Cap蛋白虽然均属于DuCV-1b亚型,但SDJN/0723、SDLC/0311与代表株DQ100076位于同一分支,SDHZ/1021、SDENJ/0429位于另一分支,共有11处氨基酸突变;4株病毒的Rep蛋白较保守,氨基酸序列与代表株参考序列之间差异较小。研究表明,DuCV-1b亚型DuCV是2022—2023年山东地区鸭群中的优势流行基因型,并且病毒基因持续变异,结果为山东地区DuCV的防控提供了参考。
In order to understand the prevalence of duck circovirus(DuCV)in Shandong region and explore its molecular characteristics,samples were collected from suspected infected ducks in some areas of Shandong province for pathogen detection,and it was preliminarily determined to be duck circovirus infection.The whole genome sequences of four DuCV strains were amplified using conventional PCR methods,and their genetic evolution and molecular characteristics were analyzed.The results showed the whole genome sequences of the four viruses ranged from 96.6%to 99.6%,and were at the same branch as the representative strain DQ100076,with a homology of over 97%,belonging to the DuCV-1b subtype.The Cap proteins of the four strains belonged to the DuCV-1b subtype,SDJN/0723 and SDLC/0311 were at the same branch as the representative strain DQ100076,while SDHZ/1021 and SDENJ/0429 were at the other branch;There were 11 amino acid mutations in the Cap protein.The Rep proteins of the 4 strains were relatively conserved with no mutations in the amino acid sequences,and there was little difference in amino acid sequence between them and the representative strain.It′s indicated that the DuCV-1b subtype of duck circovirus was the dominant prevalent genotype in duck populations in Shandong region from 2022 to 2023,with continuous genetic variation of the virus,which provided reference for the prevention and control of DuCV in Shandong region.
