基于UHPLC-Q-Orbitrap HRMS和生物活性比较分析栀子不同部位的差异

Differences of Different Parts of Gardenia jasminoides Ellis Based on UHPLC-Q-Orbitrap HRMS and Biological Activity Comparison

摘要 采用UHPLC-Q-Orbitrap HRMS方法对栀子果实、花、叶、根四个部位的谱图进行综合分析,以阐明其化学成分差异,结合体外抗炎和抗增殖活性实验来探讨栀子不同部位的药用潜力。首先,从栀子的四个部位共鉴定到53个化合物,包括环烯醚萜苷类、萜类、多酚类、黄酮类、有机酸酯类等化合物;其次,采用韦恩图和热图可视化分析,展示栀子四个部位化学成分的分布和含量差异;最后,采用LPS诱导的RAW 264.7细胞抗炎模型和PDGF-BB诱导的VSMC细胞抗增殖模型评价栀子四个不同部位的生物活性。结果表明:栀子果实、花、叶、根的化学成分存在明显差异,果实中西红花苷等二萜类化合物是独有的,与其他部位相比,栀子果实和花中成分种类和含量相近;综合活性实验,栀子花抗炎和抗增殖活性优于传统药用部位。 The UHPLC-Q-Orbitrap HRMS method was used to comprehensively analyze the mass chromatogram of four parts of Gardenia jasminoides Ellis-fruit,flowers,leaves,and roots-to elucidate their chemical composition differences.This analysis was combined with in vitro anti-inflammatory and anti-VSMC proliferation activity experiments to explore their medicinal potential.First,a total of 53 compounds were identified from the four parts,including iridoid glycosides,terpenoids,polyphenols,flavonoids,organic acid esters,and other compounds.Second,the distribution and content differences of chemical components in the four parts of G.jasminoides Ellis were displayed through visualization analysis using Venn plot and heatmaps.Finally,the pharmacological activity of extracts from the four parts was evaluated based on their effects on LPS-induced NO production in RAW 264.7 cells and PDGF-BB-induced VSMC proliferation.The results showed significant differences in the chemical composition of the fruits,flowers,leaves,and roots.The diterpenoid compounds,such as crocin glycosides in the fruits,are unique.Compared with other parts,the types and contents of components in the fruit and flowers of G.jasminoides Ellis are similar.Through comprehensive activity experiments,the anti-inflammatory and anti-VSMC activities of the G.jasminoides flower were found to be superior to those of the traditional medicinal parts.
